中文摘要 | 本試驗首先自Aspergillus amstelodami, A. candidus, A. chevalieri, A. flavus, A. fumigatus, A. nidulans, A. ochraceus, A. parasiticus, A. versicolor等九株產毒 菌株中選取六株菌絲抗原交叉反應較低者(A. candidus, A. fumigatus, A. nidulans, A. ochraceus, A. parasiticus, A. versicolor)。其次,以這六株菌之菌絲,致敏紐西蘭大白 兔製備多株抗體,並建立三明治式ELISA(Enzyme-linked immunosorbent assay),以期用於 快速檢測米及玉米中Aspergillus屬產毒黴菌。結果顯示白兔經六週致敏後所得之多株抗 體,對這九株產毒黴菌均具高度反應活性。在模擬檢測米及玉米中Aspergillus菌絲時,發 現穀物成分會降低ELISA吸光值,但在穀物濃度為4%時干擾並不明顯,免疫檢測之靈敏度可 達10μg/mL乾菌絲濃度此外,將穀物中黴菌菌數之平板菌落檢測值與穀物中菌絲量之ELISA 吸光值做線性迴歸分析時,發現此二種檢測方法之相關係數(r□)大於0.83。 |
英文摘要 | Based on the antigenic characters of Aspergillus spp., mycelial antigens of six strains (A. candidus, A. fumigatus, A. nidulans, A. ochraceus, A. parasiticus, and A. versicolor) were mixed and used to immunize two New Zealand female rabbits for the production of common antibody which were then used to establish a double sandwich enzyme-linked immunosorbent assay (ELISA) in order to detect toxigenic Aspergillus species in rice and corn. Using the ELISA, the antibodies reacted highly with all of the nine strains tested (A. amstelodami, A. candidus, A. chevalieri, A. flavus, A. fumigatus, A. nidulans, A. ochraceus, A. parasiticus, and A. versicolor). When ELISA was used to detect mycelium in rice and corn, it was found that the absorbance values obtained in the presence cereal homogenate were lower than those obtained in the absence of cereal homogenate, indicating the constituents of the cereal may interfere the ELISA system. However, this interference was not significant for 4% cereal homogenate, and the sensitivity of the assay could reach 10μg dried mycelium/mL. The correlation coefficient (r□) between the colony count of mold and the ELISA values of mycelium was over 0.83. |
