中文摘要 | 以Aspergillus parasiticus、Penicillium citrinum及Fusarium oxysporum等 三株產毒黴菌之菌絲體抗原來免疫紐西蘭大白兔,以生產抗A.parasiticus、P.citrinum 及F.oxysporum之多株抗體A1、P1、F1,並用以建立三明治式酵素結合免疫分析法( enzyme-linked immunosorbent assay,ELISA) ,分析米及玉米樣品中黴菌之菌絲量,以 評估穀物樣品受黴菌污染之程度。發現米及玉米樣品中之成分,會使菌絲之ELISA吸光值 ,比其在Tween 20磷酸鹽緩衝液(PBST)中之吸光值下降。雖然如此,ELISA吸光值和黴菌 菌絲濃度間仍有正相關,且ELISA分析應用於檢測米及玉米中之黴菌菌絲時,其靈敏度仍 維持在1μg/mL乾菌絲濃度。此外,當穀物受黴菌污染時,ELISA法比菌落計數法可更快速 地檢驗出黴菌之污染。 |
英文摘要 | Mycelial antigens of Aspergillus parasiticus, penicillium citrinum and Fusarium oxysporum were used to immunize New Zealand rabbits for the production of polyclonal antibodies A1, P1, and F1 against A. parasiticus, P. citrinum and F. oxysporum, respectively. These antibodies were used to establish sandwich enzyme-linked immunosorbent assays. When these established ELISAs were used to detect molds in rice and corn, the components of the rice and corn slightly decreased the absorbance values of the mycelium sample. However, the absorbance of ELISA still had good correlation with the concentration of mold mycelium, and the sensitivity of the assay reached 1μg/mL. When the ELISA method was compared with plate count method, it was found that the ELISA method could rapidly detect fungal contamination. |
