中文摘要 | 以化學法測定黴菌菌絲中之幾丁質可用於估算穀物受黴菌污染之程度。本試驗首 先以鹽酸水解Aspergillus parasiticus, Penicillium citrinum, Fusarium oxysporum等 三株產毒黴菌之菌絲,以便將菌絲中之幾丁質分解成葡萄胺糖,再藉比色法加以定量。其次 探討影響菌絲中葡萄胺糖含量之因子。結果顯示幾丁質之酸水解水解率平均為94.5%,在玉 米及米中菌絲之回收率因受菌種之影響而為66-93%。此外,米及玉米成分也會干擾葡萄胺糖 之測定。培養基之種類、培養溫度、培養時間及培養方式,對菌絲及葡萄胺糖含量均具有顯 著性的相互影響。 |
英文摘要 | The measurement of glucosamine content in fungal mycelium using a chemical method had been employed to evaluate fungal contamination in cereals. In this study, chitin containing mycelium of three toxigenic strains, Aspergillus parasiticus, Penicillium citrinum, and Fusarium oxysporum, was hydrolyzed by hydrochloric acid, and the glucosamine formed was then determined by using the colorimetuic method. In addition, factors which affected the content of glucosamine in fungal mycelium were investigated. The results showed that the average acidic hydrolysis ratio of chit in was 94.5%, and that the recovery of spiked mycelium from rice and corn was 66-93%, depending on the strain tested. Further, the components of rice and corn also interferred with the measurement of glucosamine. Finally, it was found that the glucosamine content of fungal mycelium was varied by the kind of cultural media, the inculbation temperature, incubation time, and cultivation method. |
