中文摘要 | 人類大腸中乙醯轉移酵素利用acetyl Co A乙醯化芳香胺類致癌物的能力是引發大腸直腸癌非常重要的危險因子。 本研究利用國人正常大腸組織經碎取純化後利用2-aminofluorene(2-AF)及p-aminobenzoic acid來檢測此酵素的活性,結果發現此酵素的分子量為34 kDa,最適宜的反應催化溫度為37℃,最適宜的反應酸鹼pH值為7.5,動能分析此純化的酵素,以2-AF為受質,其Km為0.924mM而V max為2.568nmol/min/mg protein。0.25 mM iodoacetamide會抑制此酵素約50%活性,當濃度升高到1 mM時則抑制力高達90%。檢測單、雙價離子對此酵素的影響,發現銅及鋅離子具有最強的抑制作用。這研究是第一次有關國人正常大腸組織中乙醯轉移酵素的純化及特性的報告。 |
英文摘要 | Acetylator genotype-dependent expression by human colon normal tissue for arylamine N-acetylation capacity, catalyzed by acetyl coenzyme A-dependent N-acetyltransferase(NAT), may be an important risk factor in the initiation of colorectal cancer. Human colon normal tissue cytosols from surgical samples was investigated for NAT activity toward p-aminobenzoic acid and the arylamine carcinogen 2-aminofluorene. The enzyme had a molecular mass of 34 kDa. The optimal temperature for the enzyme activity was 37℃ and a pH of 7.5 for both substrates tested. The Km and Vmax values for 2-aminofluorene were determined to be 0.924 mM and 2.568 nmol/min/mg protein, respectively. The N-acetyltransferase activity was inhibited by iodoacetamide at 0.25 mM, and activity was reduced 50%. At 1.0 mM iodoacetamide activity was inhibited more than 90%. Among a series of divalent cations and salts, Cu�杠� and Zn�杠� were demonstrated to be the most potent inhibitors. This is the first demonstration of acetyl CoA: arylamine N-acetyltransferase activity in Chinese human colon normal tissue. |
