英文摘要 | A microbore liquid chromatographic assay for the simultaneous measurement of haloperidol and reduced haloperidol in plasma is established. A deproteination procedure is applied in the pretreatment of plasma samples prior to the sensitive microbore liquid chromatographic assay. Sixteen plasma samples are readily prepared within 20min. Many advantages of the micro assay are demonstrated. Due to signal enhancement of microbore liquid chromatography, the detection limit of the present assay is 2-8pg per injection. In addition, isocratic separation of haloperidol and reduced haloperidol is achieved within 6 min. Hence, theoretically, over 240 samples can be analyzed in a working day. The required volume of a plasma sample is only 50 ul. Several aspects of the present assay are considered, including analytical linearity, precision, detection limit, and application. This rapid and sensitive microbore chromatographic assay in plasma samples are both required for the routine clinical or basic research in biological psychiatry. |
