中文摘要 | 本研究乃在了解不同亞硝酸鈉濃度在30及4℃以及不同pH(5.0~8.0)下對Aeromonas hydrophila之抗菌活性並就菌體受傷害及復甦之情形進行探討。 結果顯示在30℃及pH 5.0下,亞硝酸鈉所呈現之抗菌活性最強。培養液中50ppm之亞硝酸鈉即造成試驗菌株之完全死滅,而在5℃時則僅對試驗菌株呈部分殺滅之效果。此外亞硝酸鈉之抗菌活性隨著其濃度之減少及pH之增加而呈減少之趨勢。 在30℃下A. hydrophila 13018於含50ppm之BHIB(pH 5.0)中培養12小時後其存活菌體受傷害之比率即達100%,而在5℃下則需36小時才達類似之程度。受傷害之菌體在BHIA中大約經過9小時後其復甦率幾乎達100%。 |
英文摘要 | This study was conducted to understand the antimicrobial activity of various concentration of sodium nitrite against Aeromonas hydrophila at various pH (5.0~8.0) and at two temperatures (5℃ and 30℃). Besides, changes in the population of injured cells in the presence of sodium nitrite and the resuscitation of injured cells were also examined. Results revealed that presence of sodium nitrite in Brain heart infusion broth (BHIB) retarded the growth of the inoculated Aeromonas hydrophila cells or partially or completely inactivated them depending on the concentration of sodium nitrite, pH of broth and incubation temperature. At pH 5.0 and 30℃, sodium nitrite exhibited the greatest antimicrobial activity against the test organisms. Under this test condition, presence of 50ppm sodium nitrite in (BHIB) resulted in the complete inactivation of inoculated A. hydrophila. While only partial inactivation of test organisms was noted at 5℃. In addition, the antimicrobial activity of sodium nitrite decreased as its concentration decreased or pH of the broth increased. In BHIB (pH 5.0) containing 50 ppm sodium nitrite, the viable cells of A. hydrophila were found to be 100% injured after 12hr of incubation at 30℃. On the other hand, same degree of injury of the viable cells was noted at a longer period of incubation of 36 hr at 5℃. The nitrite-injured bacteria were found to be near 100% recovery after 9 hr of resuscitation on BHIA. |
