中文摘要 | 利用栽培種花生(Arachis hypogaea)臺南選9 號(TNS9)及臺南11號(TN11)之未熟 胚軸及子葉之五種不同部位為培植體,分別為 (1) 切除胚根及胚芽之胚軸 (代號 A); (2) 保留頂端之上半部子葉 (代號 B);(3) 保留基部之下半部子葉 (代號 C);(4) 切除頂端之 上半部子葉 (代號 B-);(5) 切除基部之下半部子葉 (代號 C-);以 MS 為基礎培養基,分 別添加 25mg/l BA(C1), 25mg/l BA+1mg/l NAA(C2) 及 2mg/l 2,4-D+1mg/l kinetin (C3) 等三種培養基,癒合組織繼代於以 MS 為基礎培養基添加 1mg/l BA+0.4mg/l NAA+1mg/l kinetin,分別添加 8g/l Difco agar(D1-a),及 4g/l 凝膠 (D1-G) 的培養基。進行花生 未熟胚軸及子葉不同部位之器官形成之誘導, 結果如下:癒合組織的誘導率,C3 培養基優 於 C2、C1 培養基。而癒合組織的量,則以 C2 培養基最多,且質地最佳。不同部位之間, 未熟子葉的癒合組織誘導率高於未熟胚軸,呈極顯著差異。五種不同部位培植體中,C 部位 培植體培養於三種培養基中皆可直接誘導芽體形成,C2 培養基的誘導率最高,C3 培養基最 低。而癒合組織繼代於分化培養基,除未熟胚軸部位 (A) 少數產生芽體 (體胚 ) 分化外, 其餘部位均無分化。 |
英文摘要 | This study attempted to compare the in vitro organogenesis on different explants of peanut cultured on different media components. Immature embryo axes and cotyledons from immature seeds of peanut cultivars TNS9 and TN11 were classified into five explant types, including embryo axis without radicle and epicoty(A), the half distal cotyledon with tip part(B), the half proximal cotyledon with base part(C), the half distal cotyledon without tip part(B-), and the half proximal cotyledon without base part(C-). The C1,C2 and C3 media used for organogenesis were prepared by adding 25mg/l BA, 25mg/l BA+1mg/l NAA, 2mg/l 2, 4-D+0.5mg/l kinetin to the MS basal medium, respectively. Results are summarized as follows:The rate of callus formation in C3 medium was much higher than those in C1 and C2. However, C2 medium was the best and C3 the worst for callus formation. The rate of callus induction from immature cotyledon was significantly higher than that from immature embryo. Among the five types of explants, only from the half proximal cotyledon with the present of the base part (C) could shoot be induced directly. The shoot formation rat in C2 medium was the highest and that in C3 medium was the lowest among the three organogenesis media. |
