中文摘要 | 本試驗以落花生栽培種臺南11號未熟胚之上胚芽為培植體,探討細胞分裂素誘體導多芽體,及生長素與抗壞血酸促進根系發育的效果,期望建立直接經過器官形成再生植株之方法。比較thidiazuron(TDZ)及N6-benzyladenine(BA)對多芽體的誘導效果顯示,添加TDZ培養基有較高的芽體形成率及芽體數,TDZ誘導多芽體及增殖的最適濃度為4mg/l~8mg/l。BA可誘導較正常的芽體,TDZ則可誘導葉片較肥厚短小之叢生芽,在芽體基部或葉腋處也著生許多芽原基。將芽體分割進行發根試驗的結果顯示,naphthaleneacetic acid(NAA)誘導根系的效果優於indolebutyric acid(IBA),添加50mg/l抗壞血酸和0.5mg/l NAA之培養基則有更子的效果,約有57.5%的芽體可再生成具有旺盛根系的植株,經移植至土壤中也可生長健旺及正常開花。 |
英文摘要 | In this study, the peanut (Arachis hypogaea. L.) cultivar TN11 was used as materials. The explants containing epicotyle excised from immature embryo were cultured on the MS basal medium supplemented with N6-benzyladenine (BA) or thidiazuron (TDZ) to find the efficient protocol for regeneration of plantlet via organogenesis. When immature epicotyle have cultured on MS basal medium with different concentration of TDZ or BA, addition of TDZ was found to be better than BA in the rate and number of shoot formation. The optimal concentration of TDZ to induce and multiply shoot was at 4 mg/l-8mg/l. Observations for morphology of shoot induced with TDZ and BA indicated that BA induce normal shoot, but TDZ induce the small and thickened multiple shoot with bud primodia at the base of stem and axil. The induced rate of rooting with naphthaleneacetic acid (NAA) was higher than indolebutyric acid (IBA). The medium supplemented with 50 mg/l ascorbic acid and 0.5 mg/l NAA could improve the growth and development of root system. 57.5% of shoots could develop to the well-rooted plantlets and develop into healthy and fertile plants when planted in soil. |
