| 中文摘要 | 本研究以栽培種花生台南選9號(TNS9)及台南11號(TN11)為材料、取不 同生育期之未熟胚為培植體、以L2為基礎培養基,分別添加 2mg/1NAA+0.01mg/1kinetin+2mg/1ADE(代號 A1);2mg/1NAA+1mg/1BA(A2);0.02mg/1PIC+40mg/1ADE十0.05mg/1 ABA(A3);另以 MSB為基礎培養基,添加2.5mg/1 2,4-D(A4)及以MS基礎培養基,添加1mg/1 2,4-D(A5):1mg/l 2,4-D+1mg/1NAA(A6)等六種培養基,藉以探討適合末熟胚培養體 胚形成的培養基、以及不同生育期對體胚形成之影響,結果如下: 不同生育期的未熟胚誘導體胚形成率以R5期最佳,無論是在何種培養基下,R5 期均較R6期有較高的體胚形成率,換言之,若以外觀而論,即為莢果長、寬發 育完全,但殼上網紋尚未形成之時。而癒合組織的誘導。則R5、R6期均佳。至 於品種間對於體胚誘導的影響,TNS9的體胚形成率高於TN11。 就六種培養基而言,A2培養基對體胚形成效果最佳,產生多量且正常的體胚, 並進而發育成芽體:A3培養基的形成率雖高,但大多為不正常體胚,無法發育成 正常芽體;而A1培養基朋有利於根的形成,產生多量粗且長的根;A4培養基能誘導 出多量的癒合組織,亦可形成體胚,而A5及A6癒合組織形成率高,但形成量不 及A4,可直接誘導體胚形成。 |
| 英文摘要 | The objective of the present study is to investigate the effect of medium composition onsomatic embryogenesis from immature embryos of peanut differing in pod development stage.Two cultivars, Tainan Selected 9 (TNS9) and Tainan 11 (TN11), and six media were used inthe experiment. Using L2 medium as the basis, three media that denoted by A 1, A2 and A3,were prepared with the addition of (1) 2 mg/1 NAA + 0.01 mg/l Kinetin + 2 mg/1 ADE, (2)2 mg/1 NAA + 1 mg/1 BA, and (3) 0.02 mg/1 PIC + 40 mg/1 ADE + 0.05 mg/1 ABA, respectively. An A4 medium was also prepared by adding 2.5 mg/1 2,4-D into the MSB basalmedium. The media A5 and A6 were prepared by using MS medium as the basis and adding1 mg/1 2,4-D and 1 mg/l 2,4-D + 1 mg/1 NAA, respectively. The results are summarized asfollows: The inducation rates of somatic embryo depended upon the pod development stages. Thesomatic embryogenesis frequency of immature embryos from pods at R5 stage was higher thanthat from pods at R6 stage. As to callus induction rate, however, R6 stage was better thanR5. Among the six media used in this study, the A2 medium was the best one, which induced many normal somatic embryos that could differentiate for shoot as well. A3 mediuminduced abnormal somatic embryos, while Al medium was good for root formation. As for A4 medium it induced a great amount of callus and some somatic embryos. A5 and A6medium had a high frequency of callus induction, since they induced somatic embryogenesisdirectly. |
