中文摘要 | 為了充分瞭解蛋白質的作用機制,有關其詳細三維空間構造之知識是不可或缺 的。目前晶體X光繞射分析,已成為研究蛋白質結構之主要方法。在此我們以大腸桿菌毒 素之免疫蛋白(ImmE7)之研究為例子,來介紹蛋白質晶體學。其中大致上有五個步驟, 1)蛋白質製備純化與結晶、(2)繞射點強度測定、(3)相角之計算、(4)模型之建立與再修 正、以及(5)結構之分析解釋。在範例中,我們將ImmE7的兩種晶體,長方晶體( orthorhombic)及單斜晶體(monoclinic),分別以多重同型置換法(multiple isomorphous replacement)及分子轉換法(molecular replacement),測定出它們的結構。 |
英文摘要 | To fully understand the functioning mechanism of a protein, detailed knowledge about its three-dimensional structure is essential. The X- ray diffraction method is one of the most important techniques for protein structure determination. Here we present a brief introduction of protein crystallography including protein purification and crystallization, data collection and processing, phase determination, model building and refinement, and structure analysis and interpretation. We also describe our work of colicin E7 immunity protein (ImmE7) as an example. ImmE7 was crystallized in two different unit cells, orthorhombic and monoclinic, and the structures were solved by multiple isomorphous replacement and molecular replacement methods, respectively. |
