中文摘要 | 最近游離基與過氧化脂質被發現與各種化學藥物引起的肝傷害有關,並且是病毒 性肝炎、脂肪肝、缺血或休克後的肝傷害之誘發或增惡因子。因此,能消除游離基與過氧化 脂質之抗氧化劑被期待為治療肝疾患的藥物。另一方面,很多治療肝病的單味中藥被發現其 作用機轉主要與抗氧化活性有密切的關係。然而中藥方劑的抗氧化作用之研究卻少有報告。 本研究目的乃是探討肝疾患常用中藥方劑之抗氧化活性。我們對 12 個中藥方劑,探討它們 的消除 superoxide (O2-) 之活性, 及抑制 CCl4 或 FeCl2 引起肝細胞脂質過氧化之效力 。進一步評估它們抑制四氯化碳誘發大白鼠肝臟細胞質過氧化之作用。結果發現,十二種中 藥方劑皆對 superoxide 有一定清除作用,且對 Fe2C12 所誘發肝細胞過氧化脂質之產生有 明顯抑制作用。另一方面,動物實驗顯示,十二個方劑中有十個方劑明顯地抑制 CCl4 誘發 大白鼠急性肝炎及脂質過氧化。此脂質過氧化的抑制作用與其肝功能改善程度成正比。十二 個方劑中以茵陳蒿湯、大柴胡湯及龍膽瀉肝湯之抗氧化作用最為明顯。此三者在臨床上常被 用於急性肝炎之治療,因此抗氧化作用可能為中藥治療急性肝炎之重要機轉,其重要性值得 進一步探討。各個單味保肝中藥的抗氧化作用也值得進一步研究。 |
英文摘要 | Recently, free radicals and lipid peroxide are found to be related to the liver damage induced by various chemical agents. Moreover, they are also considered as triggering factors or exerberation factors for viral hepatitis, fatty liver, ishemia or shock induced liver injury. Therefore, the antioxidants with scavenging effect on free radicals and lipid peroxide are considered to be as useful drugs for liver disease. On the other hand, many Chinese herb drugs used for liver disease have been found that their pharmacologic effects are due to anti- oxidation activities. However, there are rare reports concerning about the anti-oxidation effects of Chinese herb drug prescriptions. The aim of this study is to investigate the anti- oxidation effect of Chinese herb drug prescriptions which are used in treatment of liver disease. We investigated the scavenging effect of twelve kinds of herb drug prescriptions on superoxide and their inhibitory activities on the lipid peroxidation induced by FeC12. Furthermore, we evaluated their suppressing activities on the lipid peroxidation of liver cells induced by CC14 in vive. The results showed that all of the herb drug prescriptions have significant scavenging effect on superoxide induced by xanthine-xanthine oxidase. They also have significantly inhibitory activities on the production of lipid peroxide from rat -liver cells induced by FeC12 in vitro. On the other hand, in animal study, ten of twelve herb drug Prescriptions show evidently inhibitory activities on the acute hepatitis and lipid peroxidation induced by CC14. Their inhibitory activities on lipid peroxidation were correlated with the improved rate of liver function. Among twelve herb drug prescriptions, Inchenhau-tang, Tachaihu-Tang, and Rondansegan-Tang have most great antioxidative effects. These three herb drug prescriptions are mostly used in treatment of acute hepatitis. These results suggest that anti-oxidative effect may have an important role for Chinese herb drug prescriptions in the treatment of acute hepatitis. It is worthful to further investigate the antioxidative effects of each herb drug which is used in treatment of hepatitis. |
