中文摘要 | 本試驗在探討以幾丁質水解菌,幾丁質?和鹽酸水解幾丁質生產幾丁寡酗之條件。幾丁質以 Ateromonas caviae 或 K9 號菌在 30℃水解 2~10 天,其主要水解產物為幾丁二糖(GlcNAc)2 佔 64~86%,其次為乙醯葡萄糖胺 (GlcNAc) 佔 13~37%,若以市售幾丁質?Sm (EC.3.2.1.14 來自 Serratia marcescens) 或 Sg (EC 3.2.1.14 來自 Strep-tomyces griseus)在40℃水解 2 hr,其主要水解產物:Sm 為 GlcNAc 佔77~92%,Sg為 (GlcNAc)2 佔 87~90%。上述水解條件,在水解液中均未檢測到幾丁五糖 (GlcNAc)5和幾丁六糖 (GlcNAc)6,但檢測到少許幾丁三糖 (約4.0~6.4%)。幾丁質若以 10 或 12 N鹽酸在 40℃水解 1.5 hr,其主要水解產物亦為 GlcNAc 佔 57%,而 (GlcNAc)5和(GlcNAc)6 約佔5%,若以 12N 鹽酸在 50℃水解 0.5hr,其總幾丁寡醣、(GlcNAc)5和(GlcNAc)6 的製成率可高達67.4、16.1和10.3%。若水解的鹽酸濃度在 6N 以下,則水解液中檢測不到 (GlcNAc)5和(GlcNAC)6。 |
英文摘要 | The research was conducted to investigate the conditions to prepare N-ace-tylchitooligosaccharides from chitin by chitinolytic bacteria, chitinase or hydrochloric acid. Around 64~86% of N-acetylchitobiose (GIcNAc) 2 and 13~37% N-acetylglucosamine (GlcNAc) were produced from chitin by the hydrolysis of Aeromonas caviae or strain K 9 at 30℃ for 2~10 days. Hydrolysis of chitin by Sm (EC. 3.2. 1.14 from Serratia marcescens) or Sg (EC 3.2. I. 14 from Streptomyces griseus) was carried our at 40℃ for 2 hr, producing 77~92% GlcNAc or 87~90% (GIcNAc)2 by Sm or Sg, respectively. However, N-acetylchitopentaose (GlcNAc)5 and N-acetylchitohexaose (GlcNAc)6 were not detected by above hydrolytic conditions, except small amount of (GlcNAc)3 (4.0~6.4,%). About 5% (GIcNAc)5 and (GlcNAc)6 were obtained from hydrolysis of chitin by 10 or 12N HC1 at 40℃ for 1.5 hr, but about 57% GIcNAc was obtained in this hydrolytic condition. A high yield of N-acetylchitooligosaccharides from the hydrolyosis of chitin by 12N HC1 was harvested at 50℃ for 0.5hr. The yield of total (GIcNAc)2~6, (GlcNAc)5 and (GlcNAc)6 were 67.4, 16.1 and 10.57/o, respectively. Nevertheless, (GIcNAc)5 and (GIcNAc)6 were not detected in the hydrolytic liquid when the concentration of HCI lower than 6N. |
