中文摘要 | 酸酪乳(yogurt)中活乳酸菌數為重要品質指標之一,傳統平板計數法一般需要二天。本研究探討利用 Malthus 2000 微生物檢測系統 (Malthus Instruments Limited,Cra-wley,England) 應用於酸酪乳中乳酸菌菌敷快速測定的可行性。利用該儀器測得酸酪乳之電導檢測時間 (conductance derection time) 和傳統方法所測得之菌數相關性良好,其線性迴歸係數介於─0.95與─0.98之間。若酸酪乳中含有107CFU/ml之乳酸菌,則以電導法檢測僅需2小時左右,具有快速及簡便的優點,且具有自動化和分析多量樣品的能力。 |
英文摘要 | The vaible cell number of lactic acid bacteria is an important factor of the quality of yogurt. The conventional MRS plate count method for the enumeration of lactic acid bacteria in yogurt needs two days. In this study, the feasibility of a conductimetric method using Malthus 2000 (Malthus Instruments Limited, Crawley, England) system for the rapid enumeration of lactic acid bacteria in yogurt was investigated. Based on the change of conductance in the incubation ceils, the relationship between the detection time and MRS plate count was established. The linear correlation coefficients between the log value of lactic acid bacteria and the detection times were between ─0.95 and ─0.98. If the cell density of lactic acid becteria in yogurt was 107CFU/ml, the time needed for the conductimetric assay was only 2h. Conductance measurement has the advantages of rapidity, automation and high sample capacity. Therefore, the technique can be used as an alternative rapid method to the conventional plate count procedures. |
