| 中文摘要 | 臺灣泡洞之花粉發育與花序之開展有關,當花序中花芽仍處在繼續生 長狀態時,花粉母細胞、四分子及單核初期至中期均可在同一花序之花芽中觀察 到。當花序生長停止,則進入長約一個月之單核末期與有絲分裂期,然後再進入 雙核期。取花粉不同發育期之花藥培養於MS及Nistch & Nistch培養基添加不同 濃度之蔗糖(3-9%)及不同NNA與BA或NAA與Kinetin 組合濃度。結果顯示在 MS培養基添加3mg/1NAA,5mg/1BA及3%蔗糖可誘導 60%的單核期花藥形成 癒傷組織,唯此癒傷組織經切片證實為由花藥組織而來。 |
| 英文摘要 | The pollen development of P. taiwaniana has higher relationship with the development of flower buds in the a panicle. When most flower buds in panicle remain growing, the pollen mother cell, tetrad and the 1st to 3rd stage of pollen will be detected. Once the panicle stops developing, all anthers are in developing from 4th stage to early binucleate for about 1 month. Then all flower buds develop into binucleate stage and keep to the next year until the flower bloom. Based on the development of anther, every stage of anther was cultured on MS and Nistch & Nistch media adding sucrose (3-9%) and different combinations of NAA and BA or Kinetin. The result showed that the anther from the 1st to 3rd stage of pollen cultured on MS media adding 3 mg/1 NAA, 5 mg/1 BA and 3% sucrose could be induced callus mostly (about 60% of cultured anther). But under the detection of microscope revealed the callus was from the interior tissues of anther not from pollen. |
