中文摘要 | 利用栽培種花生(Arachis hupobaea L.)臺南選9號(TNS9)及臺南11號(TNll)與 野生種花生A. paraquariensis、 A. batizocoi、A. stenosperma、A. villosa 及 A. pusilla等7種花生之單核期花藥為培植體。以MS基礎培養基,添加2 mg/l IAA+4mg/l Kinetin為癒合組織誘導培養基,比較不同種花生及不同培養基對花藥癒合組織誘導率之 差異。再將誘導之癒合組織接植於1/2 MS+0.5mg/l BA+0.2mg/l BA+10g/l sucrose+ 2.5g/l gelrite(代號E9)及MS+1mg/l NAA+2mg/l BA+30g/l sucrose+3.8mg/l gelrite(代號Hl)兩種分化培養基,進行種間癒合組織分他能力之比較,結果如下:野生 種花生花藥癒合組織的誘導率皆較栽培種為高,且種(品種)間有顯著差異。野生種花生中 僅A.paraquariensis及A. pusilla之癒合組織在Hl及E9培養基均能分化形成芽體。而其 它野生種花生及栽培種花生之癒合組織皆無芽體形成。 |
英文摘要 | To study the callus induction from anthers of different species of Arachis, anthers at uninu-cleate stage from two A. hypogaea cultivars (TNS9 and TN11) and five wild species A. paraquariensis, A. batizocoi, A. stenosperma, A. villosa and A. pusilla were in vitro cultured on MS with an addition of 4 mg/1 IAA and 2 mg/1 kinetin. The calli induced were subcultured on differentiation media E9(l/2 MS+0.5 mg/1 IBA+0.2 mg/1 BA+10 gll sucrose+2.5 g/l gelrite) and HI (MS+1 mg/1 NAA+2 mg/l BA+30 mg/1 sucrose+3.8 mg/1 gelrite) respectively to compare the differentiation ability of calli among species. The results were summarized as follows. The callus induction rate of wild species were higher than that of cultivars. There was significant difference between species. Shoots differentiated from calli were observed on only the two wild species, A. paraquariensis and A. pusilla. in HI medium and E9 medium, respectively. |
