中文摘要 | 利用栽培種花生(Arachis hypogaea)台南11號(TN11)、台農5號(TN5)、立枝 仔(VB12)、VA CHICO 及VR34等五個花生品種之單核期花藥為培植體,探討生長調節 劑對花生花藥癒合組織誘導及小孢子發育之影響,以期建立花生花藥培養之技術,進而做為 育種之利用,其結果如下: 花生花藥癒合組織的形成率在品種、培養基以及品種與培養基之間均呈極顯著差異。以 TN5由D3培養基(MS+3mg/1 2,4-D+0.5mg/1 kinetin)所誘導的形成率93.6%最高,VR34 81.8% 次之。而TN11及VA CHICO於D 培養基(MS+2mg/1 2,4-D+0.5mg/1 kinetin)可獲得較高 的形成率。D 培養基所產的癒合組織結構緊密呈乳白至淡黃色,與一般胚性癒合組織相 近。 TN11單核期花藥接種於含2,4-D培養基7天後,小孢子可發育至具相同大小之2核及 3核細胞。14天後於含有2,4-D、1AA和NAA三種培養基均可發現部分小孢子成熟為花粉 粒,而部分成為多細胞團。經30天的培養,由石蠟切片的結果顯示,花藥壁與花藥囊間的 連絡組織(connective tissue)等體細胞會增殖形成癒合組織,而花粉細胞也會增殖突破藥囊, 並與由體細胞形成的癒合組織混雜。 |
英文摘要 | In order to establish the protocol for anther culture of peanut plant, the experiment was conducted to investigate the effects of plant growth regulators and the development of microspores of anther-derived callus. Anthers at uninucleate stage of the five peanut cultivars, TN11, TN5, VB12, VA CHICO and VR34 were used as materials. The results were summerized as follows: The induction of anther-derived callus among cultivars, media and the interaction between cultivar and media varied significantly. The highest rate of callus induction was found in TN5 and VR34 cultured on D medium (MS+3mg/1 2,4-D+0.5mg/1 kinetin)(93.6% and 81.8%, respectively) and higher rate was also obtaned in TN11 and VA CHICO cultured on D medium (MS+2mg/1 2,4-D+0.5mg/1 kinetin). The calli induced on D medium were compact and have white or light yellow color, similar to the characteristics of embryoid callus. After seven days of culture, the microspores of TN11 on media supplmented with 2,4-D were found have cells with the same size two and three nuclears. After fourteen days of culture, there formed cell group on media with 2,4-D, IAA or NAA. |
