中文摘要 | 本試驗之目的在探討Trichoderma reesei.菌體培養條件及其與纖 維素�t活性之關係.以作為纖維素�t生產之參考。T.reesei所分泌之纖維 素�t為胞外酵素;其中以羧甲基纖維素(carboxymethylcellulose,CMC)為基質 之內切葡萄糖���t或稱矮甲基纖維素�t(endoglucanase;CMCase)活性最 高.其次為濾紙水解�t(filterase)及產雙醣之纖維外切�t(cellobio- hydrolase).但缺乏纖維雙糖�t(cellobiase)。利用四種不同之碳源培養 T.reese,發現以avicel及纖維素(cellulose)為碳源,可產生較高之酵素活性, 其用量以1%效果最佳。而羧甲基纖維素及濾紙之效果則較差。最適酵素 生產之菌體培養條件.以培養槽溫度25℃及90rpm之振盪速率最適;而培養 液之最初pH值在3-6之間,經八天之培養後之pH值均下降至2.75,且酵素 產量亦佳;但當培養基之最初pH提高至7、8或9時,經培養結束時,其pH 值雖末下降至2.7,但酵素之生產卻受到抑制。 |
英文摘要 | Cultural conditions for the production of cellulase by Trichodenna rusei were investigated.The cellulolytic enzymes produced by T. msa cells are extracellular enzymes. Of those cellulolytic enzymes, endoglucanase with for carboxymethyl cellulose as substrate, ie, the carboxymethyl celluiase (CMCase), counted the most followed by filterase and cellobiohydrolase(avicelase). The activity of cellobiase was not found. Studies on the effects of carbon sourceson the cellulolytic activities showed that avice and cellulose were the most effective carbonsources and the optimal concentration of these carbon sources was about 1. Carboxymethylcellulose and filter paper were poor carbon sources on enzyme production. As for the cultureconditions, it was found that the optimal temperature was 25 ℃ and the optimal shakingspeed was 90 rpm. After 8 days culture, pH of the culture broth became 2.7-2.8 even if thebroth had initial pH values from 3 to 6. Under such condition, enzyme production was high.However, when the initial pH of the culture broth was 7, 8 and 9, production of cellulolyticenzymes would be inhibited. |
