中文摘要 | 本試驗利用小葉秋海棠無菌苗的葉片,以解剖刀將葉片切成5×7 mm的培植體,接種於以1/2 MS為基礎培養基添加不同濃度BA、kinetin、zeatin與含有或不含NAA的植物生長調節劑組合,經培養四週後,結果顯示添加2.0 mg L^(-1) zeatin和0.5 mg L^(-1) NAA的處理組合其不定芽增殖率最高,培植體壞死面積最低,為最佳之生長調節劑組合,誘導出來培植體在不含生長調節劑的1/2 MS基礎培養基中經繼代2次後可見花序抽出,即自葉片培植體培養開始,總共培養4.5-5月之後,可於瓶內正常開花。 |
英文摘要 | Adventitious shoot regeneration using in vitro lamina explant of Begonia parvula was investigated. In vitro lamina explants with 5×7 mm in size were cultured on 1/2 MS containing various concentrations of BA, kinetin, zeatin in combinations with or without NAA. After four weeks of culturing the highest adventitious shoot regeneration and the lowest necrosis rate was found in explants cultured in the treatment containing 2.0 mg L^(-1) zeatin and 0.5 mg L^(-1) NAA. In vitro flowering plantlets was obtained by consecutively twice subculturing regenerants on a medium containing only 1/2 MS basal salts without plant growth regulator for a total culturing period of 4.5-5 months. |
