中文摘要 | 授草Spiranthes sinensis (Pers.) Ames.授粉後10-12天的未成熟種子無菌播種於含花寶1號3g/L,半量MS維生素,peptone 2g/L、蔗糖10g/L及agar 8g/L的培養基中,一個月後約有70-80%的發芽率。以幼嫩實生苗為培植體,培養在以花寶1號為基本鹽類加BA 1-5mg/L的固態培養基內,4個月內可增殖4-6倍之不定芽。成熟植株去頂後以BA 3-5mg/L培養,2個月的增殖率約為3倍。不定芽分切後移至不含任何植物生長激素的培養基即可發根,在添加100ml/L椰子水的培養基內,其發根率可達100%。本研究顯示綬草可應用不定芽的方式進行增殖,以達大量繁殖之目的。 |
英文摘要 | Ten- to twelve-day-old immature seeds of Spiranthes sinensis were germinated in vitro on medium containing Hyponex #1 3g/L, half strength MS vitamins, peptone 2g/L, sucrose 10g/L and agar 8g/L. Germination rate was 70-80% after one month culture. Adventitious shoots regenerated from young seedlings were increased 4-6 times on Hyponex #1 medium with BA 1-5 mg/L in 4 months. Three fold of proliferation rate were obtained from wounded shoot tip of mature plant supplemented with BA 3-5 mg/L at two-month intervals. Adventitious shoots were separated and transferred onto rooting medium without any plant growth regulator. Rooting rate of regenerated plantlets reached 100% in addition of 100ml/L coconut water. These results suggested that adventitious shoot could be used on a large scale for micropropagation of S. sinensis. |
