中文摘要 | 本研究主要建立麗格秋海棠植株再生之系統。將麗格秋海棠‘Barkos’的葉柄及葉片,接種於含有 NAA (0.1、1.0 mg/l) 及 BA(0.1、0.5、1.0、5.0 mg/l)不同濃度組合之MS培養基。試驗結果顯示,不論以葉片或葉柄為培殖體,皆以添加1.0 mg/l NAA及 0.5 mg/l BA的MS培養基最佳,每葉片可誘導8.5個不定芽,每葉柄可誘導12.6個不定芽。其中以取自葉片基部的培殖體,其不定芽誘導率接近100%。將誘導的不定芽連同培殖體,接種於MS液體培養基,震盪培養八週,可促進不定芽發根與植株再生。將再生植株移至田間生長,皆具有正常外表形狀與開花特性。依石蠟切片觀察,麗格秋海棠葉片與葉柄誘導不定芽之形成,很可能經由器官分化的途徑再生成植株。 |
英文摘要 | Directed adventitious shoot regeneration from leaf and petiole explants of Rieger begonia (Begonia x hiemalis) were successfully developed. Leaf and petiole explants were cultured on MS medium containing NAA(0.1 or 1.0 mg/l) and BA(0.1, 0.5, 1.0 or 5.0 mg/l). The optimum medium for adventitious shoot induction was MS medium supplemented with 1.0 mg/l NAA and 0.5 mg/l BA both for leaf or petiole explants. The total number of adventitious shoots was 8.5 per leaf explants and 12.6 per petiole. The leaf explants near the petiole position were beneficial for inducted adventitious shoot. To improve the frequency of plant regeneration, the adventitious shoots with original tissues were transferred to shaking liquid MS medium for 8 weeks. After rooting and acclimatization process, plantlets were transplanted to the greenhouse. The morphological and flowering characteristics of regenerated plants were the same. Anatomical examinations of excised leaf and petiole explants revealed that adventitious shoots formed through organogenesis on the surface layer of explants. |
