中文摘要 | 本研究在瞭解Cellulomonas flavigena NTOU 1株產生幾丁質水解酵素及降解膠狀幾 丁質與蝦殼粉(60 mesh)的情形。本菌於修飾的鹽類果膠增培養產(MSPB)中受膠狀幾丁質或蝦 殼粉誘導產生乙醯葡糖胺?的最適濃度各為12%(V/V;或100ml MSPB含0.84g幾丁質)或1 %(w/v)。為獲得本菌產生最高的乙醯葡糖胺?的活性,在含膠狀幾丁質的MSPB中幾丁質的 含量比在含蝦殼粉的MSPB中的含量高過約3倍。本菌在含膠狀幾丁質或蝦殼的MSPB中培 養(30℃)會產生較大量(12-28g/l)的乙醯幾丁二糖及微量的乙醯葡糖胺。本菌能水解乙醯 幾丁二糖及三糖,但不水解四糖。因此本菌可能會產生外切型雙乙醯幾丁質水解?及乙醯葡 糖胺?。當本菌於MSPB與蝦殼一起培養(30℃,10天)時,在水解液中主要的胺基酸包括 兩胺酸、麩胺酸、白胺酸及牛磺酸。 |
英文摘要 | The investigation was conducted to survey the chitinolytic enzymes produced from Cellulomonas flavigena NTOU 1 and the degradation of colloidal chitin (CC) and of shrimp shell powder (SSP; 60 Mesh) by the bacterium. The optimum concentration of CC or SSP for inducing N-acetylglucosaminidase (GlcNAcase) from the bacterium in modified saltspectin broth (MSPB) was 12% (v/v; or 0.84 g of chitin in 100 ml of MSPB) or 1% (w/v), respectively. For obtaining the highest GlcNA case activity, the amount of chitin in CC-containing MSPB was about three folds higher than that in SSP-containing MSPB. During incubation (30℃) of C. flavigena NTOU 1 in MSPB CONTAINING cc or SSP, major amount of N,N'-diacetylchitobiose (GlcNAc)2 (12-28g/l) and minor amount of GlcNAc were detected. The bacterium could hydrolyse acetylchitobiose and acctylchitotriose, but not acctylchitoteraose. We suggest that C. flavigend NTOU 1 may produce exo-N,N'-diacetylchitohydrolase and GlcNA case. Alanine, acid, lcucine, and taurine were the major maino acids in SSP-containing MSPB, after incubation with the bacterium at 30℃ for 10 days. |
