中文摘要 | 本研究使用Ralstonia eutropha ATCC 17699為實驗菌株,氮源為限量基質,以連續式發酵(溫度26℃、pH 7.0),探討不同的稀釋速率、不同C/N比(20、30、40)及二種酸類基質(丙酸鈉與戊酸鈉)對菌體生合成HB、HV和PHBV的影響。實驗結果利用變異數分析,探討各因子的影響。稀釋速率對單位體積HB產率(volumetric productivity)具有顯著的影響,C/N比以及不同的酸類基質對單位體積HB產率則沒有顯著的影響。酸類基質對單位體積HV產率與碳源轉化HV產率(yield)都具有非常顯著的影響。對於HV的累積,戊酸鈉明顯優於丙酸鈉,可見戊酸納較易於菌體代謝途徑中被利用;C/N比對碳源轉化HB產出率具有顯著的影響,發現C/N比越高,其碳源轉化HB產出率會相對變低;試驗因子對單位體積PHBV產率則沒有顯著的影響。 |
英文摘要 | Ralstonia eutropha was cultivated in a continuous bioreactor with various dilution rates, C/N ratios (20, 30, and 40) and acid substrates (sodium propionate or sodium valerate) to explore the HB, HV and PHBV accumulation during fermentation. The results were analyzed through the ANOVA (analysis of variances) by using software, namely Statistica. Experimental results show that the higher the dilution rate is, the higher the HB productivity will be. Acid substrates have a very significant effect on both the volumetric productivity and yield of HV. Feeding sodium valerate seems better to the HV accumulation than feeding sodium propionate. This may mean that valerate is easier than propionate to be converted into an HV unit of PHBV in the bio-synthesis pathway. Experimental results also show that the yield of HB decreases, if the C/N ratio increases. All these manipulating factors have no significant effect on the productivity of PHBV. |
