中文摘要 | 本研究設計一組核酸引子 (primer),序列分別為 IT1: 5’ CGTAACAAGGTTTCC 3’和 IT2: 5’ AGTTTCTTCTCCTCC 3’,利用聚合酵素連鎖反應 (polymerase chain reaction, PCR),可以將文心蘭 ‘草莓’ 品種(Oncidium vokelanti cv. "Strawberry")的5.8S核糖體核酸 (rRNA)基因與內轉錄間隔區 (internal transcribed spacer, ITS)有效複製,複製產物長為 699 bp,定序後,利用 Blast分析與送入基因庫 (GenBank)內其他高等植物相同區域比較,發現 PCR複製的 DNA片段中,有 26 bp屬於18S rRNA基因的 3’端,而 52 bp屬於26S rRNA基因的5’端。因此,內轉錄間隔區 (ITS)實長為 621 bp,此區包含 ITS1, 5.8S rRNA基因與 ITS2等三區,其長度分別為 211 bp, 163 bp及247 bp,G+C百分比分別為 54.5, 58.3及56.3%。 |
英文摘要 | The nucleotide sequence of internal transcribed spacer (ITS) region between 18S and 26S ribosomal RNA (rRNA) genes of Oncidium vokelanti cv. ‘Strawberry’ was amplified by polymerase chain reaction (PCR). The primers for PCR, IT1: 5’CGTAACAAGGTTTCC 3’ and IT2: 5’ AGTTTCTTCTCCTCC 3’ were designed for amplification and sequencing. The length of PCR product was 699 bp. Compare the obtained sequence with the sequence of ITS from other higher plant species, showed that the cloned sequence contained 52 bp of 26S rRNA gene, ITS region and 26 bp of 18S rRNA gene. Thus, the length of ITS region was 621 bp, including 211 bp of ITS1, 163 bp of 5.8S rRNA gene and 247 bp of ITS2. The G+C contents were 54.5, 58.3, and 56.3%, respectively. |
