英文摘要 | A penicillin electrode was constructed by coupling a pH electrode and a penicillinase- immobilized membrane prepared by entrapping enzyme within gluten matrix. The use of gluten takes many advantages because it is biodegradable, harmless to the environment, cheaper and readily available. This penicillin electrode showed different responses to penicillin G solution, depending on the buffer stength, thickness of membrane and penicillin concentration. When the enzyme-immobilized membrane with a thickness of 40 μm was used, the steady-state response to increasing concentration of penicillin G buffered with 0.01 M phosphate was linear in the range of 1 to 12 mM. The response time was about 1 min. Empolying a higher buffer strength (0.025 M phosphate) could increase the upper limit of the linear calibration of penicillin to 16 mM, accompanying with the compensation of a lower sensitivity. Increasing the thickness of membrane increased the response time and reduced the sensitivity. Highly reproducibility was evident from the repeated measurements. |
