中文摘要 | 本研究利用青黴菌Penicillium simplicissimum w46生產胞外植酸�t(phytase) ,期能應用並藉以高植物種子及殼類中不溶性磷(植酸)之利用性。於三角瓶規模之最適化 植酸�t生產條件如下:在含有1.50% xylose, 1.50% (NH�瓷^�特O��, 0.05% KCI, 0.01% sodium phytate, 0.05% MgSO�瓷D7H�烙,0.001% MnSO�瓷D4H�烙,及0.001% FeSO�瓷D 7H�烙,初pH值6.5之培養基中接種4×10�瀛pores/50mL,於27℃ ,170 rpm 震盪培養至第 六天時會有最高之植酸�t產量為4.2U/mL。此粗酵素液經過純化後之分子量大小約為65KDa。 其最適作用pH值及最適作溫度分別為4.0及55℃,且具有廣泛之基質專一性;其活性會受到 二價及三價鐵離子與SDS之明顯抑制,而於金屬螯合劑EDTA存在時則無此現象。 |
英文摘要 | The optimal medium for the production of extracellular phytase by culturing penicillium simplicissimum W46 in shaking flasks consisted of 1.50% (NH��)�特O��, 0.05% KCI, 0.01% sodium phytate, 0.05% MgSO�瓷D7H�烙c, 0.001% MnSO�瓷D4H�烙 and 0.001% FeSO�瓷D 7H�烙. The optimal conditions were initial PH:6.5% inoculate size:4×10�瀛pores/50mL, incubation temperature: 27℃; shaking rate: 170 rpm. After 6 days of culture under the above conitions, the maximum production of phytase was achieved (4.2U/mL). The phytase was purified to electrophoretical homogeneity using CM-Sepharose CL-6B, Q-Sepharose Fast Flow and Sephacry1 S-200 chromatographs. The MW, optimal pH and temperature were 65 KDa, 4.0 and 55℃, respectively, The purified enzyme showed broad substrate specificity and was inhibited by Fe����,Fe ���吧nd sodium dodecyl sulfate, but not inhibited in the presence of EDTA. |
