中文摘要 | 停經後,雌激素缺乏,第一型介白質和腫瘤壞死因子大量分泌刺激造骨細胞分泌 第六型介白質( interleukin-6,IL-6 ),導致骨質明顯流失和骨量減少。 在風濕性關節 炎和骨質疏鬆症患者,血中 IL-6 的濃度有明顯增加的情形。 IL-6 可刺激骨髓中單核細胞 形成的數目進而增加破骨細胞的形成。 另外,IL-6 亦可促進骨吸收的進行。 因此, IL-6 經由加速骨吸收的進行,可造成骨量明顯減少。但 IL-6 是否會同時經由影響骨生成而造成 骨量的改變,即 IL-6 對骨生成的影響和機轉為何,則仍不清楚。本研究旨在探討 IL-6 對 骨生成的實際影響。 其方法為自懷孕 13 天 BALB ╱ C 母鼠取得胎鼠頭蓋骨組織,然後使 用第一型膠原蛋白�t分解法獲得單一細胞,並植入 35 mm 培養皿中培養。 實驗組培養皿加 入 IL-6 ( 0.01,0.1,1 或 10 ng ╱ ml )而對照組則只加入 BGJb 培養液。 實驗過程 中,利用位相差鏡檢、von Kossa 染色法、骨群體形成計量法、放射性氫三標定胸腺嘧啶嵌 入法、鹼性磷酸�t活性測定法和膠原蛋白合成測定法等評估 IL-6 對骨生成的影響。實驗結 果發現 IL-6 ( 1 及 10 ng ╱ ml )明顯減少骨群體形成的數目及放射性氫三標定胸腺嘧 啶之嵌入。 另外,IL-6 ( 10ng ╱ ml )明顯降低造骨細胞鹼性磷酸�t活性,但對第一型 膠原蛋白的合成則沒有影響。本研究結果顯示第六型介白質明顯抑制骨生成進行。 |
英文摘要 | An increase in production of interleukin (IL)-1 and tumor necrosis factor during estrogen deficiency can stimulate osteoblasts to secrete IL-6. Levels of circulating IL-6 are elevated in inflammatory diseases (such as rheumatoid arthritis) and osteoporosis. In addition, IL-6 has been known to be able to stimulate hematopoiesis and then increase the possibility of osteoclast formation. However, the precise effect of IL-6 on bone formation is still unknown. The purpose of this study was to investigate the effect of IL-6 on osteogenesis using an in vitro calvarial culture system. Single calvarial cells were obtained from 13 day timed-pregnant BALB/C mice by collagenase digestion and plated into 35 mm Petri dishes. IL-6 (0.01, 0.1, 1 or 10 ng/ml) was added to the experimental dishes while only Bigger, Gwatkin, Jackson b medium was added to the control. The effect of IL-6 on bone formation was evaluated using bone colony formation assay, alkaline phosphatase activity assay, �� H-thymidine incorporation assay and collagen synthesis assay. IL-6 (1 or 10 ng/ml) significantly reduced bone colony formation and �� H-thymidine incorporation. In addition, IL-6 (10 ng/ml) significantly decreased alkaline phosphatase activity. However, IL-6 had no effect on collagen synthesis. The results of this study indicate that IL-6 significantly inhibits bone formation. |
