中文摘要 | 我們應用靈的高效相層析法,以逆相碳 18 的分離管柱( reversedphase C18 ) 及移動相為 acetonitrile:water ( 65:35,v/v, PH 3.0 以磷酸調整),來沖提洋玉蘭 以及夜合花之枝幹和葉的乙醇抽提物,並確認抽提物中是否含有厚朴酚及和厚朴酚。液相層 析分離出來結果,以紫外光檢測器 290 nm 波長偵測,以及另外一套液相層析螢光檢測器做 分析結果的再認認, 此螢光檢測器的激發波長及發射波長分別設在 304 nm 以及 412 nm。 實驗結果發現,在此分析系統下,紫外光檢測器所偵測到疑為厚朴酚及和厚朴酚的成分,經 螢光檢測器的再確認後,發現洋玉蘭以及夜合花之枝幹和葉的乙醇抽提物中,並無厚朴酚及 厚朴酚的吸收波峰存在。因此,洋玉蘭以及夜合花之枝幹和葉的部位可能不含厚朴酚及和厚 朴酚,至於其樹皮部份是否含厚朴酚及和厚朴酚,則有待進一步探討。 |
英文摘要 | A sensitive high-performance liquid chromatographic method was used to identify magnolol and honokiol from 95% ethanol and 50% ethanol extracts of the stem and levave of Magnolia grandiflora L. and Magnolia coco (Lour.) DC. Chromatographic analysis was achieved on an isocratic system consisting of a reversed-phase C18 column with a mobile phase of acetonitrile:waster (65:35, v/v PH 3.0 adjusted by orthophosphoric acid) to elute the magnolol and honokiol from herbal extracts. The detection limited of UV detection and fluorescence detection were 50 ng/ml and 1 ng/ml, respectively. The eluted magnolol and honokiol were deteted at 290 nm for UV detection. The further identification was observed at excitation and emission wavelengths of 304nm and 412nm, respectively for the optimal fluorescence response. Although UV detection showed absorpitve peaks related to the retention times of magnolol and honokiol, fluorescence detection did not show any related absorptive peaks. We concluded that there is no magnolol and honokiol in the stem and levave of magnolia grandiflora L. and Magnolia coco (Lour.)DC. |
