| 中文摘要 | 本研究試驗膳食脂肪對低密度脂蛋白脂酸組成與氧化反應之關聯,同時亦測試膳食添加維生素E對低密度脂蛋白的抗氧化效應。本研究以六組大白鼠為實驗動物,三組餵食基礎飼料,其所含油脂為10葵花油、紅花籽油(高油酸含量)或牛油。另三組飼料為為基礎飼料並添加維生素E(700mgα-tocopherol succinate/kg)。經四週的飼養後,收集白鼠的血液,分離低密度脂蛋白,然後添加銅離子以促發氧化反應。氧化產物以TEARS法及共軛雙鍵化合物法測定。低密度脂蛋白的脂酸組成、維生素E及膽固醇含量亦分別分析。實驗結果發現餵食葵花油導致低密度脂蛋白之多元不飽和脂酸含量較高,其受銅離子促發之氧化程度在最初兩小時亦較其它兩種油脂餵食組高。飼料中添加維生素E並不影響低密度脂蛋白之氧化程度但可延緩葵花油組低密度脂蛋白之氧化作用。研究結果顯示餵食高量多元不飽和脂酸之油脂增高白鼠低密度脂蛋白之氧化易受性,而維生素E添加僅對高易受性低密度脂蛋白增加保護作用。 |
| 英文摘要 | Dietary manipulation to alter the fatty acid composition of LDL and its susceptibility to the copper-catalyzed oxidation was tested in this study. Also, the effect of dietary vitamin E supplementation was examined in this system. Three groups of rats were fed a basal diet containing 10 sunflower oil,safflower oil (high oleic acid content) or beef tallow. The other three groupswere fed vitamin E-supplemented diets in which 700 mg o-tocopherol succinatewas added to I kg basal diets. Blood samples were collected after a 4-weekfeeding period. LDL was isolated and the susceptibility to oxidative modificationwas assessed by in vitro incubation with copper ions. Oxidation products weremeasured as TEARS and conjugated dienes. The a -tocopherol content, fattyacid composition, vitamin E and cholesterol levels were also determined in theLDL samples. Sunflower oil feeding resulted in higher content of PUFA in LDLand more susceptible LDL over the 2-hour incubation than the other two fats.Vitamin E supplementation did not affect the extent of LDL oxidation butretarded the formation of conjugated dienes in LDL of sunflower oil .group. Thedata denoted that dietary fat containing high amount of PUFA prompted thesusceptibility of LDL to oxidation and vitamin E supplementation was protectiveonly in these highly susceptible LDL. |
