| 中文摘要 | β-乳球蛋白為牛乳中主要的乳清蛋白,根據β-乳球蛋白基因表現序列IV上 第5863位置之點變異處,經由聚合酵素連鎖反應併合限制酵素切割圖之方法,可將之區分為 AA、AB及BB三種基因型。本研究以國內荷蘭乳牛血液樣本及進口荷蘭種公牛冷凍精液為分析 對象,以PCR增幅出262 bp之產物,再以限制酵素Hae Ⅲ對PCR產物進行切割,而完成各樣 本β-乳球蛋白基因型別之鑑定。本次實驗共分析國內四牧場158頭泌乳牛及28頭公牛之冷 凍精液樣本,結果在國內泌乳牛部分,AA型有33頭、AB型有75頭、BB型有50頭,B基因頻率 為55.4%;在進口公牛精液部分,AA型有4頭、AB型有11頭、BB型有13頭,B基因頻率為 66.1%。將公牛基因型別鑑定結果與種公牛遺傳性能評估值對照統計後顯示,基因型為AB型 (11l3±138)及BB(1067±92)之種公牛比AA型(897±101)種公牛有較高之總性能指數 (TPI)(P<0.05)。本實驗之結果可供酪農作選擇冷凍精液之參考,以提高國內乳牛β-乳球 蛋白B型基因之頻率,進而提升國內乳牛的產乳能力。 |
| 英文摘要 | β-Lactoglobulin is the major whey protein in cow's milk. Due to a point mutation occurred at position 5863 in the exon IV of β-lactoglobulin gene, different genotypes can be identified by using polymerase chain reaction (PCR) and restriction enzyme digestion. In this study, 158 cows' blood samples obtained from 4 different farms and 28 imported bulls' semen samples were analyzed. A 262 bp fragment was amplified by PCR and then digested with Hae Ⅲ. Of these 158 dairy cows, 33 cows were AA type, 75 cows were AB type and 50 cows were BB type. The B gene frequency was 55.4%. In the 28 bulls analyzed, 4 bulls were AA type , 11 bulls were AB type and 13 bulls were BB type. The B gene frequency was 66.1%. Comparison between genotypes and transmitting trait evaluaton indicated that AB type (1113±138) and BB type bulls (1067±92) had higher total performance index (TPI) than AA type bulls (897±101). The results of this study can serve as a reference for dairy farmer to select the B genotype dairy bull for artificial insemination, so as to increase the β -lactoglobulin B gene frequency, and thereby elevates the milk production of dairy cattle. |
