| 中文摘要 | lpha-乳白蛋白是牛乳中一種主要的乳清蛋白,在牛乳中,alpha-乳白蛋白的量與牛乳的產量有密切的相關,由其DNA序的不同可將之區分為A、B兩型遺傳變異體,產生AA、AB及BB三種基因型。經由乳牛產乳性狀的評估發現,帶有A基因變異的乳牛有較高的產乳量、乳蛋白量與乳脂量。因此,為提高乳牛A型基因頻率以改善牛乳的品質,本研究以國內進口荷蘭種公牛冷凍精液為分析的對象,利PCR-RFLPs的方法鑑定alpha-乳白蛋白基因型別,並比較基因型與種公牛遺傳性能的相關性。實驗共分析45頭種公牛精液,AA型的結果有2頭,AB型有18頭,BB型有25頭,A型基因頻率為24.4%,與種公牛遺傳性能評估值相比,顯示AA型與AB型種公牛在產乳量、乳蛋白量與乳脂量的遺傳預測方面,有較BB型為高的趨勢。本研究結果將可供酪農篩選出A型基因的種公牛進行人工配種,增加乳牛alpha-乳白蛋白A型基因的表現頻率,進而提高乳牛泌乳的能力。 |
| 英文摘要 | Alpha-Lactalbumin is a whey protein and which is associated with the yield for milk, Regulation of this protein can be categorized by difference in DNA sequence into two genetic variants; A and B, and three genotypes; AA, AB and BB have been observed. By the trait evaluation, it was found that cattle with A gene have higher yields of milk, protein and fat. Therefore, the objective of this study using PCR-RFLPs is to analyze the alpha-lactalbumin genotypes in Holstein bulls for evaluating the A gene frequency to improve the milk yield. The association between genotypes and transmitting trait of dairy bulls was also compared. In the genotyping of 45 dairy bulls, the genotypes identified were 2 AA, 18 AB, and 25 BB. The A gene frequency was found to be 24.4%. Comparison between genotypes and stransmitting trait evaluation indicated that bulls with AA type and AB type gene have higher predicted transmitting ability for the yield of milk, protein and fat that those with BB type gene. The results of this study can serve as a reference for dairy farmer to select the A genotype dairy bull for artificial insemination, so as to increase the alpha-lactalbumin A gene frequency, and thereby elevates the milk production of cattle. |
