| 中文摘要 | kappa-酪蛋白為牛乳中酪蛋白的一種,根據kappa-酪蛋白基因exon IV相對於第148胺基酸位置的DNA變異點,利用聚合酵素連鎖反應併合限制酵素切割圖(PCR-RFLPs)的方法,可將之區分為AA、AB及BB三種基因型,帶B型基因的乳牛所產的乳,有較高的乳蛋白量與較佳的乳酪液量及品質。為提高乳牛kappa-酪蛋白B型基因的頻率,本研究以進口之荷蘭種公牛冷凍精液為分析對象,利用PCR複製出350 bp的DNA產物,再以限制酵素對變異點進行辨識切割。實驗共分析43頭種公牛的冷凍精液,結果AA型的有35頭,AB型有7頭,BB型僅有1頭,B基因頻率為10.5%,與國外分析的結果比較,有明顯偏低的現象。另外,基因型分析的結果與種公牛遺傳性能評估值相比顯示,AB型基因的種公牛較AA型種公牛有較高產乳量、乳脂量及乳蛋白量的遺傳能力預測值。本實驗的結果可供酪農作為選擇冷凍精液的參考,以提高國內乳牛kappa-酪蛋白B型基因的頻率,進而改善牛乳的品質。 |
| 英文摘要 | Kappa-casein is an important component of casein micelle. In the exon IV of bovine kappa-casein gene, sequence differences were found at corresponding to the 148th amino acid position. By using polymerase chain reaction (PCR) and restriction enzyme digestion, three genotypes, AA, AB and BB, were identified. Dairy cows with B gene have higher average milk protein yield and their milk have a better cheese quality. In order to increase the kappa-casein gene frequency, this study analyzed imported frozen semen. A 350 base pair fragment was amplified and digested with restriction enzyme. In the 43 bull semen analyzed, 35 bulls and an AA genotype, 7 bulls and had an AB genotype, and only one bull had a BB genotype. The B gene frequency is only 10.5%. When compared with other studies, the result is obviously lower. Comparison between genotypes and transmitting trait evaluation indicated that AB type bulls have higher predicted transmitting ability for milk yield, fat yield, and protein yield. The results of this study can serve as a reference for dairy farmer to select the B genotype dairy bull for artificial insemination, so as to increase the kappa-casein B gene frequency, and thereby elevates the milk production of dairy cattle. |
