中文摘要 | 本研究利用層析樣品前處理之固相萃取(solid phase extraction, SPE)常用的小層析管Sep-Pak cartridge進行膽固醇及膽固醇酯之區分,並分別測定區分物中膽固醇之含量。本研究首先於模式系統中建立區分方法,再應用於豬油中。本實驗所建立的分析方法為:將?油樣品0.1g加1ml的正己烷溶解後,注入預先以4ml正己烷潤濕過的silica Sep-Pak cartridge中,先以8ml含2%乙酸乙酯的正己烷溶液沖提,再以4ml含10%乙酸乙酯的正己烷溶液沖提之,最後更換為含20%乙酸乙酯的正己烷溶液沖提8ml。將以2%乙酸乙酯的正己烷熔液沖提所得之部分合併為第一個區分,此為膽固醇酯存在的區分,其餘溶液沖提所得之部分合併為第二個區分,此為游離態膽固醇存在的區分。將此二區分濃縮至乾,加入5α-cholestane為內標準品,經皂化後以GC測定膽固醇含量及計算膽固醇與膽固醇酯之相對比例。當以此方法來分析模式系統及外添不同量之膽固醇酯及膽固醇的市售精製?油時,不論是在總膽固醇、膽固醇酯及膽固醇之回收量上,或是膽固醇酯佔總膽固醇含量百分比上,實驗測定值皆為樣品值的90%以上。?油未外添膽固醇或膽固醇酯時其總膽固醇含量為49.3mg/100g,其中游離膽固醇為33.4mg/100g,而膽固醇酯為15.9mg/100g。利用本研究所建立之silica Sep-Pak cartridge進行樣品中膽固醇與膽固醇酯之區分具有區分時間短(10分鐘),溶劑用量少(20~30ml),樣品負載量大(0.1g油脂)及適用範圍大(膽固醇酯與膽固醇之相對百分比可由25至100%)等優點。 |
英文摘要 | A method developed for the separation and determination of esterified and free cholesterol in lard using prepacked silica Sep-Pak cartridge (solid phase extraction) is described. The sample solution (0.1g lard dissolved in 1 ml hexane) was applied to a silica Sep-Pak cartridge which had been preyiously conditioned with 4 ml of hexane. The Sep-Pak cartridge was first eluted with 8 ml of hexane/ethyl acetate (98:2, v/v), followed by 4 ml of hexane/ethyl acetate (90:10, v/v) and finally 8 ml of hexane/ethyl acetate (80:20, v/v). The hexane/ ethyl acetate (98:2, v/v) eluate contained cholesterol ester and the combined eluate of the other eluting solvents (90:10 and 80:20, hexane/ethyl acetate) contained cholesterol as confirmed by both TLC and GC analyses. The eluates were evaporated to dryness under nitrogen, they were then saponified and their cholesterol contents were determined by GC analysis. When this method was employed to measure the free and esterified cholesterol contents of model system containing standard compounds and real system (lard), the recorveries were above 90%. The total, free and esterified cholesterol contents of lard was 49.3, 33.4 and 15.9 mg/100g, respectively, the cholesterol ester content was about 32.2% of the total cholesterol in lard. The Sep-Pak separation method developed in this study had the following advantages: shorter separation method developed in this study had the following advantages: shorter separation time (10 min), less solvent used (20~30 ml), higher separation capacity (0.1g oil) and wider application range (cholesterol ester/total cholesterol from 25 to 100%). |
