中文摘要 | 許多的報告指出,以不同的定量方法來決定相當同樣品的膽固醇含量,在結果上會產生差益;而在決定膽固醇的含量時,不同的萃取溶液也會產生不一致的結果。所以這個實驗的目的是評估以不同種類和不同比例的萃取溶液對測量蛋中膽固醇之含量的影響,同時對分析蛋中膽固醇的兩種不同定量方法也在此實驗中加以研究比較。 三組蛋雞連續七十天餵予下列三種飼料:(1)飼料中添加1.5%玉米油,(2)飼料中添1.5%鯡魚油,(3)不添加任何油脂。雞蛋每天被收集記錄後,蛋黃和蛋白被分離開。混合的蛋黃在分開加入氯仿╱甲醇(67:33,v:v)和二氯甲烷╱甲醇(90:10,v:v)後,分別以氣相層析法和比色測定法(Redul & Morris,1973)加以定量分析蛋黃中膽固醇的含量。結果顯示,不同的定量方法會呈現不同的兩條曲線,但是曲線的變化卻很類似。 在研究萃取溶劑的實驗裡,蛋黃以七種不同比例的氯仿╱甲醇溶劑(100:0,90:10,67:33,50:50,30:70,10:90和0:100,v:v)萃取脂肪和膽固醇,結果發現最有效的萃取溶劑的比例是90:10。當溶劑被換成是二氯甲烷╱甲醇的實驗時,最有效萃取蛋黃中的脂肪和膽固醇50:50。實驗的資料顯示,在三種萃取溶劑50:50,67:33和90:10比例下,二氯甲烷╱甲醇能比氯仿╱甲醇萃取出更多的蛋黃膽固醇。 |
英文摘要 | Inconsistencies exist in reported values for egg yolk cholesterol due to variation in extraction solvents and quantitation methodologies. Studies were conducted to evaluate the effectiveness of different solvents and to study different methods of analyzing the extracted materials. Initially, hens were fed for 70 days on diets with no oil added, 1.5% corn oil, and 1.5% menhaden oil. Egg yolks were pooled on a daily basis from each treatment group. Yolk samples were extracted by two solvents, chloroform/methanol (67:33, v:v) and dichloromethane/methanol (90:10, v:v). The saponified extracts were analyzed for the cholesterol content by the colorimetric (Rudel & Morris, 1973) and gas chromatographic methods. Significantly different results but similar cholesterol change patterns between these two methods were found in eggs from each treatment. Egg yolk samples were extracted with seven combinations of chloroform/methanol, ranging from ratio of 100:0 to 0:100, to determine the total lipids and cholesterol content. The cost effective combination was 90:10. In the second trial, seven combinations of dichloromethane/methanol, ranging from ratio of 100:0 to 0:100, were used. The most effective ratio was 50:50. Data indicated that dichloromethane/methanol at the ratio of 50:50, 67:33, and 90:10 extracted more cholesterol from the yolk than the same combinations of chloroform/methanol. |
