中文摘要 | 儲藏性蛋白質為穀類作物種子主要的成份大分子,其組成分子的複雜性可被利用為品種辨識與品質檢定的分子標幟。除化學含量分析與電泳技術外,Reversed-Phase High Performance Liquid Chromatography (RP-HPLC)技術近年來也開始用於禾穀及豆類儲藏性蛋白質之檢測。本試驗之目的為建立一檢測稻米儲藏性蛋白質prolamins與glutelins的RP-HPLC流程,並利用三個本地栽培品種及其心腹白與透明米粒評估RP-HPLC在本省稻米品種及品質檢識上的潛力。 結果顯示,HPLC管柱溫度可明顯影響圖譜解析度,50℃較30℃(室溫)呈現較高的解析度。移動相流速也可調解圖譜解析度,1.5mL/min比1mL/min有較高的解析度。本實驗使用之條件為:管柱溫度prolamins為30℃、glutelins為50℃,流速1.5mL/min,移動相為acetonitrile,梯度於27min內由34%升至85%。 Prolamins的圖譜顯示,臺中秈10號心腹白與透明米圖譜間有明顯差異,prolamins之RP-HPLC圖譜也可明確分辨三個品種間透明米之差異,但prolamins之RP-HPLC圖譜無法明顯分辨越光米之心腹白與透明米。Glutelins的圖譜可明確分辨臺中秈10號與越光米之心腹白與透明米,也可明確分辨三個品種間透明米之差異,Glutelins圖譜之辨識能力較prolamins圖譜佳。 本試驗初步顯示,RP-HPLC可能具有辨識稻米(白米)品種與品質之潛力,本實驗室正以更多的品種確認此技術之實用性。 |
英文摘要 | Reversed phase high performance liquid chromatography (RP-HPLC) has been recently utilized in characterization and analyzing variations of seed proteins. The RP-HPLC detected variation also has been used in genotype identification and seed quality characterization. The objectives of current research have been to develop a RP-HPLC process and explore the potential of using this technique for local (Taiwan) genotype identification and quality characterization in milled rice grains. Two japonica varieties, Koshihikari and Tainung 67, and one indicavariety, Taichung Sen 10, were primarily used in the experiment. Koshihikari and Tainung Sen 10 have been recognized to have high taste quality. In column temperature test, temperature of 50°C significantly enhanced peak number and resolution of chromatogram than did the 30°C. Within the RT value between 10 and 22 min, in which most of peaks came out, increased flow rate from 1 mL/min to 1.5mL/min could apparently improve the resolution of chromatogram of prolamines. The presently used HPLC program is: 50°C column temperature, 210 nm OD detection, eluting column with gradient acetonitrile concentration from 35% to 85% step linearly in 27 min, flow rate 1.5mL/min for glutelins and 1.0mL/min for prolamines. Using described HPLC program, chromatogram of prolamins showed significant difference between white-belly and translucent rice grain in a tested indica variety. Chromatogram of glutelins showed apparent difference between white-belly and translucent grains of one japonica and one indica cultivars. Significant variations in peak number and peak RT value also appeared among three tested genotypes. Current results reveal that RP-HPLC has the potential to be applied in both genotype identification and quality characterization of milled rice grains. |
