| 中文摘要 | 為了比較嗜酸乳酸桿菌Lactobacillus acidophilus LA-1及ADH對人體乳糖利用的影響,本實驗為12位患有乳糖不適的試驗者準備了四組試驗組牛乳(含L.acidophilus活菌非醱酵牛乳,在牛乳中分別添加LA-1及ADH冷凍濃縮或新鮮培養種菌)及一組對照組牛乳(2低脂牛乳)。由於在腸胃道中未被分解吸收的乳糖,進入大腸後,會被大腸中的微生物醱酵分解產生氫氣,因此氫氣濃度可作為乳糖不適程度的指標。試驗者在分別喝下五組牛乳後8小時內的呼出氣體經收集,分析氣體中所含氫氣濃度,以了解乳糖消化不良的情形,並記錄乳糖不適症狀的程度。結果顯示,四組試驗組的呼出氫氣濃度較對照組降低了30至60,而且四組試驗組的不適症狀也較對照組明顯的緩和許多。而比較試驗組之間的效果,LA-1較ADH顯著地降低了呼出氣體中的氫氣濃度,但症狀並 無顯著的程度差異。從對菌種特性的研究,我們發現,雖然ADH的β-半乳糖背醃活性較低,但由於ADH對膽鹽的耐受性較弱,也許有助於細胞內β-半乳糖苦醣的釋出,而相對提高幫助乳糖分解的能力。另外由培養基中含乳糖的研究,顯示LA-1及ADH在含有4乳糖的MRS培養基中,具有較高的β-半乳糖���t活性。 |
| 英文摘要 | In vivo effect of LactobacUlus acidophilus LA-1 and ADH on lactose digestion was investigated. Four test meals (non-fermented acidophilus milks preparedfrom concentrated frozen cultures and fresh cultures of L. acidophilus LA-1 and ADH) and one control meal (2 low-fat milk) were fed to twelve lactosemaldigesters. Lactose maldigestion was monitored by measuring breath hydrogenexcretion at hourly intervals for 8 hours following ingestion of 400 ml of eachmeal. Intolerance symptoms produced from consumption of control milk and acidophilus milks were also recorded. The hydrogen production was 30 to 60 lower than control values for the acidophilus milks. All subjects experienced lesssevere symptoms following consumption of acidophilus milks. Althoughacidophilus milks containing strain LA-1 resulted in less breath hydrogenproduced than did acidophilus milks containing ADH, there were no apparentdifferences among symptom scores from these acidophilus milks. The higher bilesensitivity might compensate for the lower β-galactosidase (EC 3.2.1.23; β-gal)activity of ADH in its in vivo lactose digestion. Both strains LA-1 and ADH showed much higher β-gal activity when grown in MRS broth modified by replacing dextrose with 4) lactose. |
