中文摘要 | 本研究藉由幾丁聚醣(chitosan, CS)與三聚磷酸鈉(sodium tripolyphosphate, TPP)之間所形成的離子交聯(ionic-crosslinking)反應來包覆並製備分離大豆蛋白質水解物(isolated soy protein hydrolysate, ISPH,分子量:10~30 kDa)奈米顆粒。探討ISPH濃度(1~9 mg/mL)、CS分子量(475.5、231.4及131.1 kDa)及CS/TPP重量比(2/1~6/1)對奈米顆粒之特性(包括包覆率、裝載率、產率、粒徑大小及表面電荷)及其在模擬胃、腸液中安定性之情形。結果顯示,在包覆率方面,ISPH濃度愈低、CS/TPP重量比為3/1及4/1時可獲得較高之包覆率。裝載率方面,ISPH濃度及CS/TPP重量比愈高,裝載率愈高。產率方面,CS/TPP重量比愈低,產率愈高。粒徑方面,ISPH濃度及CS分子量愈低,顆粒粒徑愈小。表面電位方面,顆粒表面皆帶正電荷,而當CS/TPP重量比減少至2/1時,顆粒表面電位會出現驟降之情形(由+47.6 mV減至+24.0 mV)。在模擬胃腸液中安定性方面,CS分子量愈高、CS/TPP重量比愈低,安定性愈佳;而以高分子量之CS, CS/TPP重量比3/1,ISPH濃度5 mg/mL所製備之分離大豆蛋白質水解物奈米顆粒具有較佳之安定性。 |
英文摘要 | Isolated soy protein hydrolysate (ISPH) nanoparticles were prepared by ionic-crosslinking of chitosan (CS) with sodium tripolyphosphate (TPP). Influential factors such as chitosan molecular weight (475.5, 231.5 and 131.1 kDa), ISPH concentration (1~9 mg/mL) and CS/TPP mass ratio (2/1~6/1) on the encapsulation efficiency (EE), loading capacity (LC), process yield (PY), particle size, zeta potential and stability in simulated gastrointestinal fluids of the nanoparticles were investigated. As a result, a high EE was obtained at the low ISPH concentration and CS/TPP mass ratio at 3/1 and 4/1. LC increased in increasing ISPH concentration and CS/TPP mass ratio. A high PY was obtained at the low CS/TPP mass ratio. The particle size decreased as CS Mw and ISPH concentration decreased. All nanoparticles showed positively charged, but when CS/TPP mass ratio at 2/1, the zeta potential will reduced from +47.6 to +24.0 mV. The stability of the CS-ISPH-TPP nanoparticles in simulated gastrointestinal fluids increased in increasing CS MW and decreasing CS/TPP mass ratio. The nanoparticles showed a comparable stability in case of preparing under CS Mw at 475.5 kDa, CS/TPP mass ratio at 3/1 and ISPH concentration at 5 mg/mL. |
