中文摘要 | 針對Klebsiella oxytoca CF154所產胞外黏性物質凍乾粉末(lyophilized extracellularmucilage, LEM)進行一系列的毒性試驗,做為未來LEM應用於食品上安全之評估。使用Wistar rats為實驗動物,測試LEM之急性和亞急性動物試驗;同時應用Ames氏偵測法,利用Salmonella typhimurium測試LEM之潛在變異性試驗;再者,以雄性ICR老鼠測試 LEM之micronucleus test。在急性和亞急性之動物試驗中,Wistar大鼠飼料中添加LEM之濃度由0.1%增至3.0% (w/w),結果與對照組比較,顯示並無顯著之毒性反應(p< 0.05)。由Ames氏偵測法結果,顯示LEM之濃度達3.0% (w/w)時,不管是否有代謝活化物(S-9 mixture)存在下,對Sal. typhimurium strains TA98及TA100均無化學變異性。micronucleus test亦無明顯之陽性反應。在12週之動物試驗,於動物飼料中分別添加1.0%,2.0%,3.0% (w/w) LEM,結果並無顯示有誘發性之毒性反應。故本研究結果顯示由K. oxytoca CF154所產LEM在濃度3.0% (w/w)以下沒有毒性反應,建議可應用於食品。 |
英文摘要 | The lyophilized extracellular mucilage (LEM) extracted from Kebsiella oxytoca CF154 was subjected to a series of toxicological tests to evaluate its safety for use in various foods in the future. LEM was examined for acute and subchronic oral toxicity by using Wistar rats, and for mutagenic potential by using the Ames test using Salmonella typhimurium, and the micronucleus test by using male ICR mice. No evidence was obtained showing acute oral toxicity at a LEM concentration from 0.1% to 3.0% (w/w) in this test, nor was there any mutagenicity exhibited up to a concentration of 3.0% with and without metabolic activation (S-9 mixture). The micronucleus test showed no obvious positive response either. Using LEM as a dietary mixture at levels of 1.0%, 2.0%, and 3.0% (w/w) for feeding the rats for 12 weeks did not show any inducible signs of toxicity. The result of this study indicates that there is a lack of toxicity and supports the use of LEM extract from K. oxytoca CF154 under a concentration of 3.0% or less for a variety of foods. |
