中文摘要 | 本文探討無患子(Sapindus mukorossi)假種皮中所具有之多種生物活性。本論文以乾燥假種皮粉末(193 g)為材料,依序利用甲醇、乙酸乙酯、正丁醇及水萃取並行減壓乾燥後,其乙酸乙酯、正丁醇及水抽出物回收率分別為21.8、56.4及10.4 g。以Ames test檢測顯示其乙酸乙酯抽出物對鼠傷寒沙門桿菌(Salmonella typhimurium)TA98不具毒性,但對TA100具毒性。其水抽出物於TA98系統中,濃度達1 mg/plate時,即具毒性。無患子甲醇抽出物對間接致突變劑(aflatoxin B1)之抗致突變能力較直接致突變劑(NQNO)強,尤其對aflatoxin B1具有極高的抗致突變能力。無患子甲醇、乙酸乙酯、正丁醇及水抽出物皆能有效抑制過氧化氫誘導人類肝細胞脂質過氧化效應。綜合試驗結果顯示,無患子抽出物對於發展為抗致突變與抗氧化之保健原料具有相當大的潛力。 |
英文摘要 | Various bioactivities and traditional applications derived from pericarps of "Sapindus mukorossi" have been demonstrated interest. In this study, 193 grams of dried pericarp powder were used as the source material. The powder was extracted with methanol (MeOH) extacts were then conducted with Liquid-Liquid partition. And the ethyl acetate (EtOAc), 1-butanol (n-BuOH) and water layer subfraction were obtained. After vacuum rotary evaporation of the extracts and subfraction for EtOAc, n-BuOH and water were 21.8 grams, 56.4 grams, and 10.4 grams, respectively. After subjecting those extracts to Ames test at 1 mg/plate, EtOAC soluble fraction was cytotoxic to "Salmonella typhimurium" TA100 while was non-toxic to "S. typhimurium" TA98. However, the water soluble fraction was cytotoxic to "S. typhimurium" TA98 at the same concentration level. MeOH extract exhibited the highest antimutagenic activity against aflatoxin B_1. Further more, MeOH, EtOAc, n-BuOH and water extracts all showed inhibitory activities on lipid peroxidation in H_2O_2-induced human liver cell system. Based on the observations that extracts of S. mukorossi performed potent antimutagenic and antiperoxidative activities, further investigations on the utilization the extracts for high-valued product are needed. |
