中文摘要 | 食品在加熱、烘乾及煙燻過程中會產生多環芳香族碳氫化合物(polycyclic aromatic hydrocarbons, PAHs),部分PAHs已証實其可能會致癌,其中苯芘可作為食品中具可能致癌性PAHs存在及其效應之指標。歐盟執委會208/2005號規定,規範特定食品中苯芘最大限量,其中食用油脂為2.0 μg/kg。本研究建立了食品油脂中苯芘含量之檢驗方法。取食用油脂以isohexane/t-butyldimethylether(95 : 5, v/v)溶解後,經HR-P固相萃取匣淨化,最後以高效液相層析儀附螢光檢出器進行苯芘含量之檢測,並輔以氣相層析質譜儀進行確認。高效液相層析部份,係以Supelcosil LC-PAH層析管柱進行分離,以乙腈及水作為移動相進行梯度沖提,於螢光檢出器激發波長290 nm及放射波長410 nm偵測,回收率在80%以上,最低檢出限量為0.02 μg/kg,最低定量極限為0.06 mg/kg,本檢驗方法均符合歐盟對方法之要求。應用本研究所建立之方法檢測市售食用油脂橄欖油等20件,結果苯芘含量範圍為0.08∼0.77 μg/kg,均遠低於歐盟規範油脂之最大限量2.0 μg/kg,顯示無安全顧慮。 |
英文摘要 | Polycyclic aromatic hydrocarbons (PAHs) form during food processing or cooking. Some PAHs were considered as carcinogens. Benzo(a)pyrene was used as a marker for the occurrence and effect of carcinogenic PAH. In Commission Regulation (EC) No 208/2005, maximun level of benzo(a)pyrene in edible oil was 2.0 μg/kg. A method to analyze benzo(a)pyrene in edible oil was developed in this study. Benzo(a)pyrene was extracted from edible oils using isohexane/ t-butyldimethylether (95:5, v/v) and cleaned up with HR-P cartridge. The extracts were analyzed by high performance liquid chromatography with fluorescence detector (HPLC/FL) and confirmed by gas chromatograph mass spectrometer (GC/MS). The Supelcosil LC-PAH column was used for the separation in HPLC. The LC was effected with a gradient elution with mobile phase of acetonitrile and water. Excitation and emission wavelengths of the detector were 290 nm and 410 nm respectively. The recoveries were all higher than 80% with the limit of detection of 0.02 μg/kg and the limit of quantification of 0.06 μg/kg. This method was well qualified for the performance criteria of analysis that is requested by EC. A survey on benzo(a)pyrene in various edible oils shows a concentration range of 0.08~0.77 μg/kg. |
