中文摘要 | 本研究針對油脂中的游離膽固醇建立一種快速而簡易的氣相層析方法,其特點是 將油脂直接分析而不必先製成衍生物,使用不分流直接注入式氣相層析儀以及非極性短管柱 DB-1(5.5 m�e0.53mm)。添加回收實驗中膽固醇之回收率介於98%至100%之間,變異係數小 於4%。皂化反應後,油脂中的總膽固醇含量亦可以本法分析測定。以本方法測定六種動物 性油脂(豬油、牛油、魷魚內臟粗油、精製魷魚內臟油、鱈魚肝油及蛋油)之總膽固醇及游 離膽固醇含量,結果顯示六種動物性油脂之總膽固醇含量依序為118 mg/100g、90mg/100g、 1252mg/100g、1218mg/100g、1647mg/100g以及3539mg/100g,而游離膽固醇含量則分別佔 其總膽固醇含量之73.8%、71.0%、19.0%、19.8%、21.4%以及84.6%。 |
英文摘要 | In this study, a rapid and simple method for determining the free cholesterol in animal fats and oils without conversion to its derivatives was developed. A splitless direct injection gas chromatograph with anon-polar short column(DB-1, 5.5 m�e0.53mm)was used. The recovery of the added cholesterol ranged from 98% to 100% with a variation coefficient of less than 4%. After saponification of the oil sample, total cholesterol could be determined using this method as well. Six samples, including lard, tallow, crude squid visceral oil, refined squid visceral oil, cod liver oil and egg oil, were quantitatively analyzed for total and free cholesterols. The total cholesterol contents of these oils and fats were 118 mg/100g, 90 mg/100g, 1252 mg/100g, 1218mg/100g, 1647 mg/100g and 3539 mg/100g, respectively. Free cholesterols were 73.8%, 71.0%, 19.0%, 19.8%, 21.4% and 84.6% of total cholesterols, respectively. This result suggests promising appliction in versatile biological samples. |
