中文摘要 | 一種未確定學名的小球形綠色藻種,經光學顯微鏡、掃描式電子顯微鏡觀察外部型態,及穿透式電子顯微鏡內部細胞胞器等構造觀察後,併同18S-28S rRNA轉錄區間報氧核醣核酸序列比對,結果認定為Chlorella Beijerinck的小球藻。此小球藻在光學顯微鏡、掃描式電子顯微鏡下觀查,呈圓形或橢圓形,約2-4μm,藻體表面平滑,和Chlorella外部形態特徵相同。穿透式電子顯微鏡顯示內部細胞胞器等也是Chlorella種類之基本構造。細胞壁可以明顯看出,三層膜結構則不易。母細胞壁有些細胞則可清楚看出。澱粉核普遍存在細胞中,外圍有大的澱粉鞘包圍,內部沒有發現類囊體薄膜層穿過,葉綠體也沒有明顯外膜包圍,整體結構、形狀和大多數小球藻Chlorella種有些不同。再利字此藻種18S-28S rRNA轉錄區間去氧核醣核酸序列和GenBank資料庫的其它藻種比對,結果顯示和Trebouxiophyceae的種類為相同群,而和Chlorophyceae則為不同群組。Trebouxiophyceae的種類含大多數的Chlorella種類,綜合上述各種分析方法所得結果,推論此一球形綠色藻種可能為Chlorella Beijerinck中一新種。由於Chlorella內部構造變異很大,分類不易,因此仍須待生化分析、更詳細的超顯微結構及分子基因配合方能確定。 |
英文摘要 | An unknown unicellular green alga, cultured in the Tungkang Marine laboratory (Biotechnology Division), Fisheries Research Institute, COA, was identified as chlorella Beijerinck species, using scanning and transmission electron microscopy and sequence comparison of ribosomal DNA between the regions of 18S-28S rRNA. Scanning electron microscopy shows that the cells of the alga are round or ellipsoidal, which are main morphological characters of Chlorella species. Transmission electron microscopy shows that ultrastructural organelles and components in the cell of this specimen are more similar to those of Chlorella species than others. The pyrenoid is common in the cell and surrounded by the large starch sheath. Its matrix is divided transversely into two hemisphere. The thylakoid lamellae are not clearly shown to penetrate the pyrenoid matrix. The cell wall of the present specimen is properly stained, and a trilaminar structure is not clearly visible. The mother cell wall remains faintly visible in some cells. The gene sequence between 18S and 28S rRNA containing 5.8S rRNA gene in internal transcribed spacer reigon in the present specimen is more closely related to those of Trebouxiophyceae species, including most of Chlorella species, than to those of Chloro-phyceae species. Thus, we conclude that this algal specimen is a chlorella species. The species definition, however, must be refined with more biochemistry and by physiological and molecular comparisons with other species. |
