| 中文摘要 | 傳染性花柳性腫瘤是狗很常見的疾病,其細胞相在組織切片中常與組織細胞瘤及 淋巴瘤之細胞非常相似,故本研究中,以組織學,免疫組織化學及穿透式電子顯微鏡觀察, 對此三種細胞的組織結構,細胞內某些抗原及超顯微結構做詳細的觀察及比較。結果發現, 傳染性花柳性腫瘤與組織細胞瘤在組織結構上細胞排列較緊密,淋巴瘤細胞排列則較鬆散。 前兩者之 Vimentin 免疫組織化學染色均為陽性,而 Keratin,Desmin,S--100,及 NSE 染 色均為陰性,淋巴瘤細胞對上列五種抗原均為陰性。電子顯微鏡觀察發現三者之超顯微結構 明顯不同, 其中傳染性花柳性腫瘤細胞含有很多胞器 (Organelles),尤其含有大量的粗內 質網,當細胞發生變性時,會擴張成空泡,此種病變在細胞抹片中即可見到,可作為診斷依 據; 組織細胞瘤的細胞雖也含有一些胞器,但其細胞核因其核外被 (Nuclear envelope) 多 處深度的內凹 (Invagination)' 造成多形性 (Pleomorphic) 或分葉現 (Lobulation), 淋 巴瘤之細胞僅含有少量的胞器。根據以上結果,可知此三種腫瘤之組織結構及細胞抗原性均 很相似,但超顯微結構明顯不同,因此,以穿透式電子顯微鏡觀察是最好的鑑別方法。 |
| 英文摘要 | Transmissible venereal tumor (TVT) is a common disease of the dog. Since its histological feature is very similar to histiocytoma and lymphoma cells, it is important to find a method for making a precise diagnosis. In this study, the TVT cells were compared with histiocytoma and lymphoma cells by histological, immunohistochemical and ultrastructural examinations.TVT cells were observed to be always arranged compactly or grew in cords and were divided into some nests by a little fibrous connective tissue. The cells contained a round or ovoid nucleus and abundant cytoplasm. Similar result was found in the histiocytoma except the nuclei were pleomorphic. On the contrary, lymphoma cells were round and contained various amounts of cytoplasm. In immunohistochemical staining, both TVT and histiocytoma cells were positive for vimentin, but negative for keratin, desmin, S-100, and neuron- specific enolase (NSE). None of the stain used were positive on the the lymphoma. In ultrastructural observation, the nucleus of TVT cells contained abundant euchromatin with one or two prominent nucleoli. The cytoplasm contained many organelles, such as mitochondria, rough endoplasmic reticulum (RER), ribosomes, Golgi complex.and lysosomes. Many microvilli protruded from the cytoplasmic membrane and interdigitated between two adjacent cells. In degenerated cells, the obvious lesions were dilatation of RER and mitochondrial swelling. On the other hand, the histiocytoma cells contained abundant cytoplasm with a moderate number of organelles, and the lymphoma cells contained little cytoplasm and only few organelles. From our results, we suggest that transmission electron microscopy used to elucidate the ultrastructural features of the three similar round cell tumors as being the best criterion for differentiating among TVT, histiocytoma, and lymphoma cells. |
