中文摘要 | 本實驗主要是探討使用 50% 酒精溶液抽取香椿葉的萃取物對 3T3-L1 脂肪細胞對[3H]-2-deoxyglucose 攝取量的影響。當細胞經過不同濃度的香椿葉萃取物(0.001、0.01 及 0.1 mg/mL)經過 60 分鐘處理後,脂肪細胞對葡萄糖的攝取量由控制組的 0.23 nmol/min/mg protein 增加到 0.30、0.33 及 0.38 的攝取量,然而在胰島素存在下,細胞對葡萄糖的攝取量會增加到 0.35,而香椿葉萃取物會更進一步使細胞對葡萄糖攝取量增加到 0.41、0.46 及 0.52 nmol/min/mg protein。若 3T3-L1 脂肪細胞預先經過 20 mM 葡萄糖處理 48 小時後,雖然細胞對葡萄糖攝取量降低,但是香椿葉萃取物也會增加細胞對葡萄糖的攝取。在胰島素存在下 cycloheximide 及 calphostin C 會抑制胰島素及香椿葉萃取物增加細胞葡萄糖糖攝取的量,但是在沒有胰島素刺激的細胞,cycloheximide 及 calphostin C 不會抑制香椿葉萃取物增加細胞葡萄糖糖攝取的活性。由以上結果顯示,有胰島素或沒有胰島素的培養液中,香椿葉萃取物都會增加 3T3-L1 脂肪細胞對葡萄糖的攝取量,但是兩者可能透過不同調節機轉。 |
英文摘要 | The effects of substances extracted from Toona sinensis leaves, using 50% alcohol/water, on cellular [3H]-2-deoxyglucose uptake in differentiated cultured 3T3-L1 adipocytes were investigated. Following treatment of cells with 0.001, 0.01, or 0.1 mg/mL extracts for 60 minutes, [3H]-2-deoxyglucose uptake increased from a basal value of 0.23 nmol/min/mg protein to 0.30, 0.33, and 0.38 nmol/min/mg protein, respectively. In insulin-stimulated cells, cellular [3H]-2-deoxyglucose uptake was enhanced by Toona sinensis leaf extract from a basal value of 0.35 nmol/min/mg protein to 0.41, 0.46, and 0.52 nmol/min/mg protein, respectively. Cellular glucose uptake was also enhanced by Toona sinensis leaf extract after incubation of cells with 20 mM glucose for 48 hours. Cellular glucose uptake with a combination of Toona sinensis leaf extract and insulin was significantly inhibited by pretreatment of cells with the protein synthesis inhibitor cycloheximide and the protein kinase C inhibitor calphostin C in normal-, medium- and high-glucose media. However, the glucose uptake-enhancing effect of Toona sinensis leaf extract was not diminished by cycloheximide and calphostin C in the absence of insulin. These results indicate that enhancement of cellular glucose uptake by Toona sinensis leaf extract in basal and insulin-stimulated 3T3-L1 adipocytes may be mediated by distinct mechanisms. |
