中文摘要 | 本研究旨在探討完整2-細胞期兔胚之細胞週期期別及不同電融合參數 (電場強度、電激次數)對家兔胚兩胚葉細胞電融合效率之影響。家兔胚之細胞週 期期別之區分,係以1-細胞期分裂形成2-細胞期後所經歷之時間為依據,分為 30 min以內(<30 min)、30~60 min、1~2h及6h以後(>6h)等四期。當應用2.5 kV ╱cm電場強度、60μs電激持續時間和2次連續電激之電融合參數電融合兩胚葉 細胞時,於<30min組之融合率(25.0%),明顯低於其餘各期別組(87.5,97.4和 89.7%,p<0.001)。將發育成2-細胞期後1~2h之家兔胚以2.0~3.5 kV╱cm不等 電場強度、60μs持續時間、2次連續電激之結果顯示,經2.0 kV╱cm電場強度 處理組之融合率(68.8%)不如應用2.5和3.0 kV╱cm之兩組者(98.2%和96.7%, p<0.01)。若以2.5 kV╱cm電場強度、60μs持續時間、1次電激處理,其兩胚葉 細胞之融合率並未較施以2次者為低(86.1%和87.1%)(p>0.05)。因之建議,電 融合完整之家兔2-細胞期胚之胚葉細胞時,應避免選取剛分裂為2-細胞期後30 min以內者,並採用2.5 kV╱cm電場強度及60μs持續時間配合1次電激之電融 合參數組合為宜。 |
英文摘要 | The purposes of this study were to evaluate the effects of cell cycle stages and electrofusion parameters on fusion rate of blastomeres in the intact 2-cell stage rabbit embryos. Cell cycle stages of embryos were classified into four stage periods(<30 min,30~60 min,1~2 hr,and>6 hr after the first cleavage). Rabbit embryos in different cell cycle stages were treated with 2 pulses of 2.5 kV/cm field strength which lasted 60 μs each. The fusion rate of the blastomeres in 2-cell embryos in early stage(<0.001). Rabbit embryos at 1~2 hr post their first cleavage were used to determine the effect of field strengths of 2.0~3.5 kV/cm. A significantly higher fusion rate was observed when the field strength was at 2.5 or 3.0 kV/cm as compared to that at 2.0kV/cm (98.2 and 96.7% vs. 68.8%, P<0.01). In addition, when the electric parameters were set at a field strength of 2.5 kV/cm and a pulse duration of 60μs for 1 or 2 pulses, the fusion rate of the blastomeres in the 2-cell embryos were the same (86.1% vs. 87.1%, P>0.05). In conclusion, the results suggested that a field strength of 2.5 kV/cm combined with one pulse of 60 μs duration were the most suitable condition to fuse the blastomeres of the 2-cell rabbit embryos electrically at a period of 1~2 hr after the first cleavage. |
