中文摘要 | 本研究旨在評估以細胞電融合技術或╱和海藻酸鈣微膠囊包埋處理或聚集胚葉細 胞之方式用於產製嵌合動物之可行性。於離體培養之結果顯示,源自小鼠二不同品系(ICR 或C578BL/6系)之2-細胞胚裸露(-ZP)之二枚胚葉細胞,被施以電融合再以海藻酸鈣微膠 囊包埋( +ME )處理後,融合胚於離體發育迄桑椹或囊胚期之比率( 42.1% )與直接以完 整之 2- 細胞胚(含透明帶,+ZP )所電融合者( 46.8% )相近; 二者均不如未電融合之 對照組者( 83.4% )( P<0.05 )。 上述融合胚,不論是在電融合後立即移置,或於離體 培育迄桑椹期或囊胚期再行移置,均未能經由偽孕處理之受胚母小鼠育成仔小鼠。當小鼠之 裸露 2 一細胞期胚經海藻酸兮微膠囊包埋後,逕行移置入受胚母小鼠之二側輸卵管內, 迄 懷孕終了,也未能育成仔鼠。資訊說明,以電融合二枚裸露胚葉細胞產製嵌合動物之不適切 性。又,電融合處理對融合胚於離體發育率之傷害性,不因事先之微囊包埋處理而有所改善 。而甲 146 枚( 73 對)源自小鼠不同毛色品系之 8- 細胞期胚,以聚集方式所產製之 71 枚嵌合胚,經離體培育迄桑椹期或囊胚期後始被移置,迄懷孕終了,育成 10 集仔小鼠;其 中,至少 6 隻具備表毛色或╱和眼睛色澤之嵌合特徵。由聚集嵌合方式育成之仔小鼠迄 30 日齡之平均體重與由完整 8- 細胞胚所育成之對照仔小鼠之差異不顯著( 18.93 vs 21.91 g,P<0.05 ); 說明由多一倍之胚葉細胞數聚集之嵌合胚所育成之仔小鼠,迄 30 日齡之體 重並未受影響。 |
英文摘要 | The purpose of this study was to evaluate the probability of producing chimeric animals by electrofusion and/or microencapsulation and aggregation techniques in early mouse blastomeres or embryos. The result indicated the proportion of the electrofused embryos with 2 blastomeres deerived individually from two 2-cell embryo collected form two different coat-color strains (ICR and C57BL/6) developed to morula or blastocyst stage (42.1%) in vitro was similar to the fused blastomere from an intact 2-cell embryos (46.8%). However, each of them was lower than the control embryos (83.4%)(P<0.05). The electrofused embryos by 2 blastomeres as indicated above and the denuded mouse 2-cell embryos embedded in Ca-alginate gel were transferred to the oviducts or uterus of synchronized recipient immediatedly popst-electrofusion or at morula or blastocyst staage developed in vitro. None of the offspring could be obtained from electrofused embryos or from denuded 2-cell embryos embedded in Ca-alginate gel. The result indicated that it was inadequate for electrofused 2 denuded blastomeres from different strain to produce chimeric mouse. Microencapsulation could not improve the in vitro development of fused embryos. However, of the 71 chimeric morulas or blastocysts produced and developed by the aggregation of two denuded embryos in 8-cell stage form two- coat-color strains (ICR and C57BL/6) in vitro were transferred to the uteryus of recipient (24 hr previous to donor), 13 live young were obtained. Of 10 pups survived, according to overt chimeric coat-color of external features and/or eye pigmentation, at least 6 were chimeras. The body weight at 30 days of age of 10 chimeric offsprings aggregated from two-8-cell embryos at 30 days of age of 10 chimeric offsprings aggregated from two 8-cell embryos were not different from the 9 pups in control group (18.93 vs 21.91 g,P<0.05). |
