中文摘要 | 本研究旨在探討13種醣類在培養基中對Aeromonas caviae之生長及水解蝦殼能力之影響。A. caviae在含1%纖維二糖、果糖、葡萄糖、麥芽糖、甘露醇、乳糖、半乳糖、蔗糖和可溶性澱粉之營養肉羹(natrient broth, NB)中,於30℃培養48小時會使培養液(pH 7.0)之pH下降至4.8左右,菌體會變長、相連在一起,失去運動性,且細胞圍有破損之現象。在這些能被A. caviae利用之醣類中,唯有含乳糖或半乳糖的培養液之pH值仍維持在5.6以上,菌體正常並具運動性。A. caviae在含1%葡萄糖之NB培養48小時後其總生菌數降低4個對數值,在培養60小時後測不出生菌數,但不含葡萄糖者之菌數仍維持在108 CFU/ml以上。草蝦殼經A. caviae於30℃水解10天後,其固形物殘留率為43%,基質中若含1%葡萄糖,則其固形物殘留率反而提高(50%),其水解能力較不合葡萄糖者差(p<0.05)。所以基質中含醣類(尤其是葡萄糖)時,不利於A. caviae的生存,且降低其水解蝦殼之能力。 |
英文摘要 | The effect of I3 carbohydrates on the growth of Aeromonas caviae and the hydrolytic ability of this bacterium on shrimp shells were studied. After cultivation at 30℃ for 48hr in nutrient brothe (NB) containiing 1% cellobiose fructose, glucose, lactose, galactose, maltose, mannitol, sucrose or soluble starch, pH of media decreased from 7.0 to about 4.8, the cells formed chain-like fiaments losing motility, and the cell envelopes were damaged. Among these carbohydrates, only galactose or lactose was utilized by A. caviae kept its motility, normal cell size, and pH of the media was kept at 5.6 or higher. In 1% glucose-containing NB, the aerobic plate count of the media decreased 4 log cycles. After cultivation for 60 hr, no viable bacteria were detected; however, the viable counts of the control (without containing glucose) remained at 108 CFU/ml or higher. When .4. caviae was cultivated in the media containing shrimp shell at 30℃ for 10 days, the solid residue (on dry basis)was 43.%; while the residue increased up to 50% when the medium contained 1% glucose. However, the shrimp shell-hydrolyzing ability of A. caviae was lower in glucose- containing medium than the medium without glucose (p<0.05). Therefore, a medium containing carbohydrate, especially glucose, was not available for the growth of A. caviae and also decreaseded the ability of the bacterium to hydro-hyze shrimp shells. |
