中文摘要 | 本研究利用牡丹酚( paeonol )來推測對大白鼠( SD )血液和肝臟 cytosols 的乙醯轉移酵素( NAT )活性的影響,此 NAT 酵素活性的測定是以 p-amino-benzoicacid 和 2-aminofluorence 作受質再利用高壓層析儀來分析被此酵素乙醯化的受質產量和未乙醯 化受質的產量, 結果顯示大白鼠血液和肝臟 cytosols 的 NAT 活性受牡丹酚的影響,且隨 著牡丹酚劑量增加,NAT 活性也上升。同時也發現該酵素動能反應常數 Km 和 Vmax 亦增加 ,由這些結果顯示牡丹酚能促進大白鼠血液和肝臟 cytosols 乙醯轉移酵素的活性。 |
英文摘要 | In this study, we investigated the effect of paeonol on the arylamine N-acetyltransferase (NAT) activity in Sprague-Dawley rat cytosols of blood and liver tissue. The NAT activity was measured by high preformance liquid chromatography for assaying the amounts of N-acetyl-2-aminofluorene and N-acetyl-p-aminobenzoic acid and remaining 2-aminofluorene and p-aminobenzoic acid. The NAT activity in the blood and liver tissue cytosol were increased by paeonol in a dose-dependent manner. The Km and Vmax of NATs were increased by paeonol in the cytosols of blood and liver tissues. From this result, we demonstrated that paeonol promoted the NAT activity in vitro in the rat blood and liver tissues. |
