中文摘要 | 本研究利用反應曲面法,探討自Aspergillus candidus CCRC 31543液體培養生產抗氧化物最適化之培養時間、攪拌速率及培養溫度等物性因子。在三因子三階次之實驗設計下,試驗之相關係數(r)值為0.9645,根據統計迴歸分析顯示,在所設定物性條件範圍內,各項因子對培養濾液乙酸乙酯萃取物回收量之影響如下:培養時間為極顯著因子(p<0.01),攪拌速率為顯著因子(p<0.05),而培養溫度與各因子間之交感項均未達顯著水準(p<0.05)。液體培養最適之物性條件分別是培養時間12天、攪拌速率80-100rpm及培養溫度26℃。最適條件下培養濾液與菌絲體之乙酸乙酯萃取物均有很強之抗氧化性,且兩種萃取物經高效能液相層析分析,其主要成分種類相似。 |
英文摘要 | Response surface methodology was applied to a shake-flask culture to determine the optimal incubation time, agitation speed and incubation temperature for the production of antioxidant from Aspergillus candidus CCRC 31543. With a designed method of three variables and three levels, the correlation coefficient (r) was determined to be 0.9645. After statistical regression, with regard to each variable for the yield of ethyl acetate extract, the incubation time and agitation speed were significant (p<0.01 and p<0.05, respectively). However, the interaction terms between temperature and other variables were not significant (p>0.05). The optimal conditions for the shake-flask culture were as follows: incubation time, 12 days; agitation speed, 80-100 rpm; and incubation temperature, 26℃. Both ethyl acetate extracts from broth filtrate and mycelia incubated under the optimal conditions exhibited high antioxidant activity. The major components of these two extracts showed similar profiles in high-performance liquid chromatograms. |
