中文摘要 | 高溫熱水解(121℃, 30 min)所生成的吳郭魚之魚皮明膠水解物(skin gelatin hydrolysate, SGH),我們發現具有清除α,α-diphenyl-β-picrylhydrazyl (DPPH•)自由基與抑制油脂自氧化的能力。為了提昇SGH抗氧化活性,本研究針對SGH製程中三個反應條件:磷酸濃度(PC)、水皮比例(WS)與高溫熱水解時間(TT)的數值,藉由反應曲面法(RSM)來探討SGH的抗氧化性最適化(optimization)。由實驗設計所生成的15種SGH,其中清除DPPH•自由基(反應濃度20 mg/ml SGH)與抑制油脂自氧化(反應濃度0.2 mg/ml SGH)最佳的表現分別為72.30%與71.25%。此數值與未最適化的最初SGH比較,經最適化生產的SGH在清除DPPH•自由基提高了22.05%,在抑制油脂自氧化方面提高14.2%。此外,由RSM預測對清除DPPH•自由基,最適化的SGH產生於PC=7.5%,WS=0.5,and TT=180 min時,其清除率為71.77%;而RSM預測對抑制油脂自氧化,最適化的SGH產生於PC=5.7%,WS=0.5,and RT=180 min,最高抑制率為72.76%。從經濟利益觀點,由最適化熱水解條件下生產的抗氧化性吳郭魚SGH,不需進一步酵素水解。因為吳郭魚是台灣的大宗養殖魚種之一,且在魚片生產過程中會產生大量廉價的魚皮副產物,此SGH具有高度潛力去開發成天然抗氧化劑。 |
英文摘要 | Skin gelatin hydrolysate (SGH) from tilapia was produced by thermal hydrolysis with a retorting treatment (at 121℃ for 30 min) and the SGH showed noticeable α, α-diphenyl-β-picrylhydrazyl (DPPH•) free radical scavenging and lipid peroxidation inhibition. To improve the antioxidant properties of SGH, three processing factors, namely acid concentration (PC), water/skin ratio (WS), and thermal hydrolysis time (TT) were then optimized using response surface methodology (RSM). The DPPH• scavenging effect (20 mg/ml SGH) and lipid peroxidation inhibition (0.2 mg/ml SGH) of the most effective sample in the RSM were 72.30% and 71.25%, respectively. Compared to the original SGH, the optimized SGH exhibited 22.05% more scavenging of DPPH• and 14.2% more inhibition of lipid peroxidation. Moreover, the highest values of the predicted DPPH• scavenging and lipid peroxidation inhibition were 71.77% (PC=7.5%, WS=0.5, and TT=180 min) and 72.76% (PC=5.7%, WS=0.5, and RT=180 min), respectively. From the economic view, an antioxidant tilapia SGH produced under an optimal condition without further enzymatic digestion has a promising potential as a natural antioxidant because tilapia is one of the most cultured fish in Taiwan and a large amount of low-priced skin byproducts can be obtained from tilapia filleting industries. |
