英文摘要 | We have established an in vitro macrophage/foam cell model to evaluate intracellular oxidation of metabolized lipids. Oxidation of low density lipoprotein (LDL) is a major cause of early atherogenesis. Massive accumulation of oxidized LDL in macrophages generates foam cells, a hallmark for early atherosclerotic lesion. Since application of antioxidants to hypercholesterolemic animals in doses sufficient to protect LDL from oxidation does not generally retard atherogenesis, cellular processes associated with cytokines might play a central role during atherogenesis. We hypothesized that intracellular oxidation of metabolized lipid might correlate to these cellular processes. In the current study, we showed that superoxide anions were produced in foam cells loaded with acetylated LDL. Overexpression of interleukin-1 beta was also observed. We concluded that intracellular oxidation in foam cells is closely associated with the generation of superoxide anions and overexpression of interleukin-1 beta. |
